Objective: Numerous clinical studies indicated that colon cancer is one of the most common alimentary diseases in men.There are no effective therapies such as surgery,chemotherapy and traditional chinese medicine.There are various factors associated with a rising risk of colon cancer and an increasing trend of mortality rates.Insidious onset,slow progress and symptoms of no specific difference make it difficult to diagnose the cancer early,therefore,most patients are found with advanced stage.Within 20 years,cytokine-induced killer cells(CIK)appeared as a new type of therapy with high tumoricidal activity and extremely low side effects.CIK kills tumor cells directly,while regulating and enhancing the body's immune function,it is an important means of a new promising approach to harness the body.CIK is one new adoptive cellular immunotherapy ivolves the transfer of immune cells that expanded and activated ex vivo.In recent years,the applications of CIK has evolved from observation into early clinical studies.CIK exhibits high proliferation rate,low cytotoxic activity in killing tumor cells in vitro and regulates immune function in vivo.Tetrandrine(Tet)are considered with a wide range of pharmacological effects such as cardiovascular and inflammatory pharmacological effect,recent researches showed Tet is also a potential anti-cancer drugs in the treatment of gastrointestinal cancer,especially colon cancer.In order to clarify the role of the jointed CIK cells with Tet on SW620 colon cancer cell lines by MTT assay.CIK cells alone or in combination with different concentrations of the Tet showed suppression of colon cancer cell line SW620,exploring a new way for the treatment of colon cancer.Methods: Collecting 12 healthy human peripheral blood samples,isolated peripheril blood mononuclear cell(PBMC)with a density gradient centrifugation and cultured in vitro.In the laboratory using sequential cytokines and monoclonal antibodies directed induction and expansion of CIK cells which were analyzed by flow cytometry.CIK cells number and phenotypic changes increased with the progress of culture in 14 days.In different concentrations of Tet liquids mixed with CIK cells and SW620 cells were determined by MTT assay,Tet group,CIK cells group and Tet joint CIK group was detected.Result: Extracting 12 healthy donors PBMC,increasing the total number of cells was detected by flow cytometry analysis,the proportion of CIK were rising in 14 days,the results showed CD3 + CD56 + cells growth from 0.13% before the onset of the experiment to 27.8%.(P<0.05)CIK cells at different effector to target ratios for colon cancer SW620 cells in vitro increased;MTT results showed that with the increase of efficiency of the target ratio,CIK cell killing activity gradually increased,and different concentrations of Tet and SW620 cells were co-cultured for 48 h,the inhibitory activity was enhanced with the increase of concentration.And in the joint group,the apoptosis of SW620 cell was significantly higher when Tet 30?mol/L joint CIK cells with effector to target ratio of 40: 1(P< 0.05).Conclusion: Tet can enhance CIK cells in vitro activity against SW620 cells and provides a new experimental basis for clinical treatment of colon cancer.CIK cells combined with traditional chinese medicine increased the pro-apoptotic effects of SW620 cells.
