Objective: To explore the expressive difference in Breast cancer susceptibility gene 1(Breast cancer susceptibility,BRCA1)and tumor metastasis suppressor gene(Non-metastasis23 NM23)between benign and malignant tumors,and to explore the expressive difference in BRCA1、NM23 between young breast cancer and elderly breast cancer,and to explore the relationship of these two genes in breast cancer that have different axillary lymph node metastasis and clinical stage.At last,to explore the relationship of these two genes in breast cancer.Methods: 91cases(including 20 patients of young breast cancer,31 patients of elderly breast cancer,20 patients of young benign breast tumor,20 patients of elderly benign breast tumor)were selected for the experiment from 2011 to 2015 in Yijishan Hospital of Wan Nan Medical College,and all of them had no treatment before surgery.All of the breast cancer patients had modified radical mastectomy surgery,and benign breast tumor patients had minimally invasive surgery and open surgery.Immunohistochemistry technique SP method was used to detect the expression of BRCA1 and NM23-H1 protein in 51 cases of breast cancer and 40 cases of benign breast tumor.Statistics software SPSS22.0 was used to calculate the result,and P <0.05 was considered significant statistical.Analyzed the expression of these two genes in breast cancer and benign breast tumor,and studied these two genes expressing in young breast cancer patients and elderly breast cancer patients.Analyzed the relationship of these two genes in breast cancer that have different axillary lymph node metastasis and clinical stage.At last analyzed these two genes that have some relationships or not.Results: 1,The positive expressions rates of BRCA1 and NM23-H1 in breast cancer were: 29.41%、31.37%,while the expressions rates of BRCA1 and NM23-H1 in benign tissue were: 62.5%、75%,the expressions rates of BRCA1 and NM23-H1 in breast cancer were lower than that in benign breast tissue(P <0.05);2,The positive expressions rates of BRCA1 in young breast cancer were: 50%,and in elderly breast cancer were: 16.13%,the rates of BRCA1 in young breast cancer were higher than that in elderly breast cancer(P <0.05);The positive expressions rates of NM23-H1 in young breast cancer(10%)were lower than in elderly breast cancer(45.16%),P <0.05;3,The positive expressions rates of BRCA1 in breast cancer with stage I-II were 34.88% which were higher than the rates of BRCA1 in breast caner with stage III(0%),P <0.05;The positive expressions rates of BRCA1 in breast cancer without lymph node metastasis were 42.31% which were higher than the rates of BRCA1 in breast cancer with lymph node metastasis(16%),P <0.05;The positive expressions rates of NM23-H1 in breast cancer with stage I-II were 37.21% which were higher than the rates of NM23-H1 in breast caner with stage III(0%),P <0.05;The positive expressions rates of NM23-H1 in breast cancer without lymph node metastasis were 50% which were higher than the rates of NM23-H1 in breast cancer with lymph node metastasis(12%),P <0.05;4,There was a positive correlations were between the expressions of BRCA1 and NM23-H1 in the breast cancer(correlation coefficient rs = 0.306,P = 0.029).Conclusions: Through this experiment,compared with benign breast tumors,the positive expressions rates of BRCA1 and NM23-H1 were lower than it.So the low expressions rates of BRCA1 and NM23-H1 may lead to breast cancer;The positive expressions rates of BRCA1 and NM23-H1 get down when it have a high clinical stage and lymph node metastasis in breast cancer,BRCA1 and NM23-H1 may play important roles in the invasion and metastasis of breast cancer;Compared to elderly breast cancer,the high positive expressions rates of BRCA1 in young breast cancer may lead to a early pathogenesis in breast cancer,and The low positive expressions rates of NM23-H1 in young breast cancer may lead to the result that young breast cancer has a high malignancy and a bad clinical stage;There is a positive correlations are between the expressions of BRCA1 and NM23-H1 in the breast cancer.To monitor these two genes may play an important role in breast cancer.
