The Effect Of JNK Signal Pathway In Gastrin Promoting Colorectal Cancer Cells Proliferation

Objective: On the basis of in vitro animal experiments,to study the proliferation and apoptosis of human colorectal cancer CACO2 cell line B which express CCKBR by the action of gastrin,and the relationship among JNK signaling pathway?gastrin and CACO2 which express CCKBR in this process.Methods : We cultured the cells in vitro.We used cell counting plate for routine counting when CACO2 cell concentration reached 1×107/ml.we established nude mouse transplanted tumor model of colorectal cancer with the CACO2 cells.We divided the mice into three groups marked A?B and C.Do not deal with A group as blank group,inject gastrin 4?g under the skin in transplanted tumor as gastrin group.As with the C group,we inject inhibition of JNK signal pathway 10?g under the skin in transplanted tumor besides injecting gastrin under the skin in transplanted tumor as inhibition of JNK signal pathway group.,Stripping each group ‘s transplanted tumors after several weeks,weight the tumor and measure the volum.in addition,detect the expressing of JNK gene's m RNA of each group's transplanted tumors through PCR technology.detect the expressing of JNK1/2 protein through Western blot technology.Result :The weight of transplanted tumor:the weight of gastrin group(7.79 ± 0.34)is more heavier than blank group(4.59 ± 0.64),(P<0.05).Gastrin group(7.79 ± 0.34)is more heavier than inhibition of JNK signal pathway group(4.91 ± 0.37),(P<0.05).The volum of transplanted tumor: Gastrin group(3.16 ± 0.14)is more bigger than blank group1.86 ± 0.26.(P<0.05).Gastrin group(3.16 ± 0.14)is more bigger than inhibition of JNK signal pathway group(1.99 ± 0.15),(P<0.05).The expression level of m RNA:.Blank group(0.913±0.097)is lower than gastrin group(2.375±0.342),(P<0.05).gastrin group(2.375±0.342)is more higher than inhibition of JNK signal pathway group(1.527±0.326),(P<0.05).The express of JNK1/2protein: Blank group(0.231±0.011)is lower than gastrin group(3.192±0.282),(P<0.05).Gastrin group(3.192±0.282)is higher than inhibition of JNK signal pathway group(0.612±0.124),(P<0.01).Conclusion:Gastrin can promote proliferation of colorectal cancer cells CACO2 which express CCKBR.The mechanism is that gastrin combine with CCKBR of colorectal cancer cells CACO2,increase the activity of JNK signal pathway.So as to promote the proliferation of colorectal cancer cells CACO2.
