Objective: To observe the various changes of bone turnover markers(BTM)in Murine calvarial model of osteolysis and response after anti-osteolysis treatment.1.Reflect the bone metabolism in the model;2.Detect and screen meaningful indicators which are suitable for early diagnosis of Al and monitor anti-osteolysis therapy outcomes;3.Better understanding the mechanisms of AL and raising awareness of the disease;4.Provide references for further clinical research and pharmaceutical research.Finally reached their early prediction,early diagnosis,early treatment purposes,to prevent or delay the development of AL as far as possible,achieve the purpose of avoiding or delaying revision surgery to improve patient function and quality of life.Methods: Titanium particle-induced mice calvarial model of steolysis.Mice were divided into 3 groups,Control group received PBS,the experiment group received 30 mg titanium particles stimulation,and the treatment group received 30 mg titanium particles +alendronate0.3 mg/(kg·w).Kill the mice after collecting blood samples postoperative 14 days,remove the mice's skull for Micro-CT scan in order to get three-dimensional images and 3D measurement analysis.Hematoxylin-Eosin Staining: observed the degree of osteolysis.Serological parameters of boneturnover markers include: TRACP-5b(tartrate-resistant acid phosphatase),RANKL(receptor activator of NF-?B ligand),OPG(osteoprotegerin),PINP(N-terminal peptide of type I procollagen),CTX(cross-linkedCtermianl telopeptide of type I collagen),ALP(alkaline phosphatase),OCN(osteocalcin)and RANKL/ OPG ratio in this animal model induced by particles were analyzed.Result: Micro-CT: Three dimensional reconstruction of the mice skull showed no significant osteolysis in the control group,particles of titanium implants led to significant reductions in bone mass,significantly reduce osteolysis in the treatment group compared with the experimental group,targeted regional BMD in the experimental group were significantly lower than those in the control group and treatment group(p<0.05),the difference was statistically significant.Serum BTMs:TRACP-5b,CTX in experimental group was significantly higher than both control group and treatment group,the treatment group was lower than experimental group but higher than control group(p<0.05),the difference was statistically significant.Serum PINP,ALP level in experimental group were significantly lower than the control group and treatment group,while treatment group was more higher than control group(p<0.05),the difference was statisticallysignificant.OPG and OPG/RANKL ratio in experimental group was lower than other two groups,The result was similary with previous studies.RANKL and OCN level in each group there was no statistical difference.(p>0.05)Conclusions:The change of serum CTX,TRACP-5b,OPG,ALP,PINP lever and RANKL/OPG ratio in accordance with osteolysis in Murine calvarial model of osteolysis,all of which can reflect osteolysis occurrence.PINP and ALP,two bone formation markers,may reflect the outcome of anti-osteolysis effectively.Combined with previous research results,we have reasons to believe that the BTMs mention above can be used for monitoring and evaluating the treatment effect of implants aseptic loosening and periprosthetical osteolysis,also can provide references for further clinical research and pharmaceutical research.Early intervention to delay or avoid revision surgery maybe come true in the near future,but need more research to be confirmed.
