ObjectiveTo explore the protective effect and possible mechanisms of Ginsenoside Rg1 combined with bone marrow mesenchymal stem cells(BMSCs)transplantation on cerebral ischemia reperfusion injury(CIR)in rats brain.MethodsOne hundred twenty Sprague-Dawley(SD)rats were randomly divided into sham-operated group,ischemia reperfusion(IR)group,IR group treated with BMSCs transplantation(IR+BMSCs),IR group treated with Ginsenoside Rg1(IR+Rg1),IR group treated with Ginsenoside Rg1 and BMSCs transplantation(IR+Rg1+BMSCs)(n=24).To establish a CIR model,left middle cerebral artery occlusion(MCAO)was used.Each group was treated accordingly in CIR 24 h.In CIR 24 h,7d,14 d,the neurological deficit was assess by modified Neurological Severity Scores(m NSS).In CIR 24 h,brain water content and TTC staining were detected to identifiy model and assess the treatment efficiency,HE Staining was used to explore the pathologic changes of ischemia lateral cortex,TUNEL was used to explore apoptosis cells of ischemia lateral cortex,Immunofluoresence was utilized to explore the levels of neuron-specific enolase(NSE)and glial fibrillary acidic protein(GFAP)of ischemia lateral cortex,Western blotting was used to explore the levels of apoptosis-related proteins such as bcl-2 and bax of ischemia lateral cortex.ResultsCompared with the sham-operated group,IR group showed significant symptoms of neurological deficit,brain edema and cerebral infarction,While compared with the model group,symptoms of neurological deficit,brain edema and cerebral infarction were significantly improved in other treatment groups,in which combined treatment group showed the best effect.IR group showed significant ischemia and hypoxia changes in brain tissue,while the changes in other treatment groups have different degrees of improvement,and combined treatment group showed the best effect.TUNEL staining found that the sham-operated group has few apoptotic cells,IR group showed obvious apoptotic cells,While compared with the model group,the number of apoptotic cells in the remaining treatment groups was decreased,and did not find apoptotic BMSCs.Immunefluorescence showed that the sham-operated group appeared many NSE and GFAP positive cells,IR group found few NSE and GFAP positive cells,While compared with the model group,the number of NSE and GFAP positive cells of the remaining treatment groups was significant increased,in which combined treatment group showed the best effect.Western blotting showed that compared with the sham-operated group,the bcl-2/bax ratio in the IR group was decreased.Compared with the IR group,the bcl-2/bax ratio of BMSCs treatment group has no obvious change,While the bcl-2/bax ratio of Ginsenoside Rg1 treatment group and combined treatment group increased significantly,but there was no significant difference between the two groups.ConclusionsGinsenoside Rg1 combined with BMSCs transplantation can promote brain tissue repair after cerebral ischemia reperfusion injury mainly by enhancing neuron like cells differentiation and the anti-apoptosis effect of Ginsenoside Rg1.
