Objective:To explore the correlation between SERT gene promoter single nucleotide(SNPs) rs25531 and rs956304 and the Cancer Related Fatigue(CRF) of colorectal cancer,also to analyze the correlation of the interaction of genetic and non-genetic factors.Methods:309 colorectal cancer patients,which select in the Second Affiliated Hospital of Nanchang University.The Chinese version of the Brief Fatigue Inventory(BFI-C) was used to evaluate the cancer-related fatigue(CRF).Collecte the peripheral blood of the patients,and extracted the genomic DNA.Direct sequencing was used todetermine the rs25531 and rs956304 genetypes,while recording the factors of gender,age, smoking, alcohol consumption, body mass index (BIM), physical activity,tumor stage, chemotherapy and radiotherapy.SPSS22.0 was used to do statistic analysis,?2 test was used for Hardy-Weinberg genetic equilibrium analysis;Logistic regression analysis was used to determine the relationship between the factors and cancer-related fatigue,calculation the adjusted OR and 95% confidence interval.All statistical tests were two-sided probability test,significance difference level were set at P<0.05.Result:The rs25531 and rs956304 genotype frequencies conform the Hardy-Weinberg genetic equilibrium(x2=0.02, P>0.05; ?2=2.89, P>0.05).Regression analysis shows the effect of cancer-related fatigue independent factor are the degree of gender, physical activity, smoking,tumor stage, chemotherapy and the genotype of rs25531.Smoking and chemotherapy is risk factors,adjusted OR are 2.41 (95% CI= 1.54?3.78) and 2.33 (95% CI= 1.45?3.75). Cancer-related fatigue is lighter in the colorectal cancer patients which are men and make physical exercise,adjusted OR are 0.58 (95%CI=0.36?0.92) and 0.37 (95%CI=0.24?0.59). Compared to the TNM stage ? patients, the risk of incidence and severity of CRF in colorectal cancer were 0.21 (95% CI= 0.06?0.32) and 0.31 (95% CI= 0.13?0.71) in stage ? and stage II patients, the difference was statistically significant, while the case of CRF in stage III patients was not statistically significant. Compared with the AA genotype, the risk of the severity of CRF increased to 2.07 times(95% CI= 1.20-3.56, P= 0.009) in the patients with rs25531 allele G carriers genotype(AG+GG genotype).Conclusion:There is no correlation between SERT gene promoter single nucleotide rs956304 and the Cancer Related Fatigue of colorectal cancer. SERT gene promoter single nucleotide rs25531 is associated with the Cancer Related Fatigue of colorectal cancer,the G variant genotype is independent risk factors against the Cancer Related Fatigue of colorectal cancer,this effect is more obvious in the patients who are women,lack of physical,smoking,TNM stage III and undergoing chemotherapy.
