Objective:Research the cholinergic anti-inflammatory pathway in astragalus polysaccharides combined ibuprofen anti-infection effect. Methods:We chooses forty healthy male SD rats, which were randomly divided into four groups and ten rates in each group(respectively labeled group A, group B, group C, group D). The four groups were using cecal ligation and puncture(CLP) rat model of sepsis. Group A for blank control group:modeling for four days before the intraperitoneal injection of nomal saline(25ml/kg, qd), after 2h and 8h nomal saline lavage modeling intervention(20ml/kg); Group B for ibuprofen group:modeling for four days before the intraperitoneal injection of nomal saline(25ml/kg, qd),2h after modeling and 8h ibuprofen lavage intervention(20mg/kg); Group C as a group of astragalus polysaccharides :modeling for four days before the intraperitoneal injection of astragalus polysaccharides(25mg/kg, qd), after 2h and 8h nomal saline lavage modeling intervention(20ml/kg); Group D for combination group:modeling for four days before the intraperitoneal injection of astragalus polysaccharides(25mg/kg, qd), after 2h and 8h ibuprofen lavage modeling intervention(20mg/kg). Prior to modeling, modeling after 2h(ibuprofen before intervention), 8h after modeling(ibuprofen twice before intervention), 12 h after modeling extract femoral venous blood 1.5 ml. Adopting micro plate method to detect the acetylcholine content in serum and ELISA method for the determination of the content of TNF-? and IL-6 in serum; Modeling 12 h after laparotomy, observe the rat peritoneal lesions, and take right upper lobe tissue was observed histopathological changes. Results:After modeling 12 h, Serum drug combination group TNF-?, IL-6 content was significantly lower than the control blank group, the ibuprofen group and astragalus polysaccharide group, there were significant differences(P <0.05). Ibuprofen group and control blank group, APS group and control blank group, the serum TNF-?, IL-6 content was significantly lower than the control group, there were significant differences(P <0.05).Rats plasma TNF-?, IL-6 agreed to establish changes in the CLP model trend began to increase 2h after modeling, show a gradual downward trend peaked in the after 8h. After modeling 12 h, serum Ach combination therapy group was significantly higher than the blank control group, the ibuprofen group and astragalus polysaccharide group, there were significant differences(P <0.05). After modeling 12 h, laparotomy, seen in the large number of purulent exudate abdominal cavity, intestinal hyperemia, expansion, adhesions, distal necrotic cecum, smell and fecal odor, on the rat lobe tissue HE staining, compared with the blank control group, ibuprofen and inflammation in the astragalus polysaccharide group to alleviate, inflammation in the combined treatment group was significantly reduced. Conclusion:Astragalus polysaccharides and ibuprofen could significantly inhibit the release of pro-inflammatory cytokines, joint use of astragalus polysaccharides and ibuprofen can produce better anti-inflammatory effect, cholinergic anti-inflammatory pathway may be involved in inflammatory processes Astragalus polysaccharides combined ibuprofen.
