| Purpose: This paper belongs to the field of TCM rheumatology.This experimental study by observation Yu Ling gout granules effect on acute gouty arthritis model rats ankle swelling degree and serum interleukin(beta(IL-1)and tumor necrosis factor(TNF-a)expression and Ibuprofen Sustained release capsule and contrast,so as to verify the jade Ling Gout Granule on anti inflammation,detumescence effect,and to explore its mechanism,to provide experimental data for clinical research on new drugs.Method: 60 Wister rats were adaptively fed for 1 week after randomly divided into blank group,model group,ibuprofen group,Yu Ling Gout Granule(high,medium and low dose of six groups,each successive drugs for 7 days,on the 7th day after dosing Coderre method will except the blank group the other 5 groups modeling,continue to give the medicine 3 days(measured during each of the time points in rats ankle swelling degree),in modeling the first 4 days of anesthetized rats,blood from the abdominal aorta,preparation of serum,enzyme linked immunosorbent assay determination the concentration of serum IL-1,TNF-?.Result:1.The changes of ankle joint swelling degree of rats in each groupThe mice in 1H groups(except blank group)showed no significant difference(P > 0.05);after the model 6h,there was a statistically significant difference between the high dose group and model group and ibuprofen group(P < 0.05),while the model group and ibuprofen group had no significant difference(P > 0.05);after modeling 12 h there were statistically significant differences between the groups,and the model group(P < 0.05),high dose group and 6h no significant difference(P < 0.05);after the model 24 h,there was a statistically significant difference between ibuprofen group and high dose group and model group(P < 0.05),but no statistically significant difference between them(P > 0.05);after the model 48 h,there was significant difference in the ibuprofen group,high dose group,middle dose group and model group(P < 0.05),there was a statistically significant difference between the low dose group and high dose group(P < 0.05).(Table 1,table 2,figure 1)2.The content of IL-1 in serum of rats in each groupYu Ling Gout Granule(high,medium and low dose group were with the rats in the model group,the serum IL-1 content have significant difference(P < 0.05),and Yu Ling gout granula high dose group and blank group rats serum IL-1 content no significant difference(P > 0.05),the ibuprofen group and model group,no significant difference(P > 0.05);different doses of Yu Ling gout granules effect on rat serum levels of IL-1 have statistically significant differences(P < 0.05),high dose group < middle dose group < low dose group < model group.(Table 3,figure 2)3.The content of TNF-in serum of rats in each groupBlank group and the other groups in rat serum TNF-a levels had a statistically significant difference(P < 0.05);model group and Yu Ling Gout Granule particles between the low dose group no significant difference(P > 0.05);model group and ibuprofen group are statistically significant difference(P < 0.05);different doses of Yu Ling gout granule group and ibuprofen group in the presence of statistically significant difference(P < 0.05);blank group < ibuprofen group < high dose group < middle dose group < low dose group(Table 3,figure 3)Conclusion:1,Yu Ling Gout Granule can improve the toes swelling of rats.2,Yu Ling Gout Granule can reduce IL-1 beta and TNF-? rat model of gouty arthritis in serum.3,Yu Ling Gout Granule in order to achieve the purpose of treatment in the treatment of gout arthritis mechanism may through to downregulate IL-1 beta and TNF alpha's levels. |
