Background After extracapsular extraction surgery or lens injury, The residual cortex or lens epithelial cells hyperplasia, the lens become turbidity, called paosterior capsular opacification. It is the most common complications after ECCE.In aqueous humor, postoperative inflammation or mechanical trauma in cataract surgery can produce plenty of the secretion and release of cytokines, these cytokines act as stimulus signal combining with the repceptors on the surface of the lens epithelial cell membrane. Lead to the cell signal transduction pathway activation.The study found that the phenomenon of posterior capsular opacification accompanied by various information transfer process between the cells. Rho/Rock signaling pathway is a basic way in eukaryotic cells. Involved in polarity, cell division, cell proliferation and apoptosis, cell adhesion and migration, ect. However, it is a pity that we now know a little between Rho/Rock signaling pathway and PCO.Objective Establish an improved model of posterior capsular opacification(PCO) in SD rats. Using western blot method and immunohistochemical method to detect the key factors Rho A, Rock?and P-MLC expression in PCO. Explore the effect of Rho/Rock signaling pathway in PCO.Methods 1.Selected 30 healthy adult SD rats. Divided them into control group(0 day) and experimental group(3day?7day?14day?28day) randomly. Each group of six. All rats under general anesthesia, All rats' right eyes were performed ECCE.Observe the Anterior segment and the development of PCO at different times. 2.The TGF-? expression in aqueous humor at different times was detected by ELISA. 3.Killed the rats and removed the right eyes for fixing?slicing?HE staining. 4.Take off the pcsterior capsular at different times. Using Immunohistochemistry and western blot to detect the expression of Rho A?Rock??P-MLC in posterior capsules.Results 1.The longer the time, the more opacity the posterior capsular was. 2.At different times HE staining showed different degrees of proliferation and migration of LECs. The longer the time, the LECs proliferation migration more significantly, more no rules. 3.After suegery, TGF-? in aqueous humor was activated, the content gradually increased.0 day was the lowest and 28 day was the highest. The difference of TGF-? was statistically significant between experimental group and control group(P<0.05). 4. Rho A?Rock??P-MLC expressed in posterior capsules at different times. The longer the time, the stronger the positive staining. 5.Using western blot to detect the expressions of P-MLC?Rho A?Rock?in posterior capsular. The expression were gradually increased. 0 day was the lowest and 28 day was the highest. The difference(the expressions of P-MLC?Rho A?Rock?in posterior capsular) was statistically significant between experimental group and control group(P<0.05).Conclusion 1.During the formation of PCO. The expression of Rho A, Rock?and P-MLC in posterior capsular gradually increased. 2.The Rho/Rock signal pathway involved in PCO formation.
