Objective: To research the influence of AMD3100 on neural stem cells proliferation and differentiation after spinal cord injury in rat,and investigate the molecular mechanism of CXCR4/?-catenin signaling.Methods: Thirty six female Sprague-Dawley rat were randomly divided into three groups: the sham group(group A),the control group(group B)and AMD3100 group(group C).Both group B and C were used to establish the model of spinal cord injury by Allen's method.The rats in group C were injectd with AMD3100 0.1mg/kg every day after SCI,while group B were injected with PBS by the same way.BBB score were performed to evaluate the hind limb function.Animals were euthanized at 7th day after surgery.All the tissue were analyzed using HE staining RT-PCR immunohistochemical method to observe the expression of SDF-1/CXCR4 axis and Wnt/?-catenin signaling.Statistical analysis of the correlation between each groups.Results: All the SCI models with paralysis of the hind limb obviously.BBB score showed significant difference between each groups(P<0.05).Observed the rats function recovery was very slow,and its degree was quite limited.RT-PCR and immunohistochemical analysis found that the expression of SDF-1,Nestin,Wnt and ?-catenin in SCI group increased significantly compared with control group,the number of ?-tubulin positive cells increased significantly,Neural stem cells proliferation and differentiation can be observed obviously.Moreover,the expression of nestin Wnt ?-catenin were decreased and the number of ?-tubulin positive cells were reduced in the group C which rats were injected with CXCR4 inhibitors.Conclusion: 1?Endogenous neural stem cells can be activated by the stimulation of SCI,and then increased neural stem cells proliferation and differentiation.2 ? The expression of SDF-1/CXCR4 axis Wnt/?-catenin signaling were increased after spinal cord injury.But treated with inhibition of CXCR4 by AMD3100,inhibitors the expression of nestin differentiation of endogenous neural stem cells via knockdown of CXCR4/ ?-catenin signaling.
