Objective:This subject is to study the expression and effect of Sirtuins proteins in variety of species normal retina,and if sirt1 could exert its neuroprotective effect against retina degeneration in animal glaucoma model,thus plays a guiding role in the diagnosis and treatment of clinical glaucoma optic nerve injury.Methods:The expression level and pattern of sirtuins protein were detected in SD rats,C57BL/6 mouse and human eyes through western blot and immunochemistry.Ischemia-reperfusion injury were performed in SD rats,Tonollab measureed baseline and high intraocular pressure,optic nerve crush were performed in C57BL/6mouse,resveratrol were injected intraperitoneally for three days from the day before IRI or ONC,the expression of sirt1 and retina struction were detected at 1 week later by western blot,HE staining and immunohistochemistry.Results:Sirt1and sirt2 protein were expressed in normal SD rats,C57 mice and human retina.Western blot showed that sirt1 protein level decreased(P ﹤ 0.01),HE staining and immunochemistry showed the struction of retina had been destroyed,the number of cells in the retinal ganglion cell layer were decreased 7d after IRI or ONC.Intraperitoneal injection of resveratrol showed a protective effect at 1 week(P﹤0.01).Conclusion:1.Sirt1 and sirt2 protein is expressed in normal SD rats,C57 BL / 6mice and human retina;2.Compared with the control eye,the expression of sirt1 declined in the retina of the experimental eye;3.Sirt1 activator resvaratrol plays a neuroprotective effect on the degenerative retina of the animal glaucoma model.
