| To investigate the effects of polysaccharide from Cyclocarya paliurus(CPP) on DNA methylation, mRNA expression and the activity of high-fat emulsion-induced hyperlipidemic rats, the hyperlipidemic rats were randomly divided into CPP high dose, middle dose, low dose groups, simvastatin, model control and control groups, each group was orally administrated with 400, 200, 100 mg/kg·BW CPP per day, water, 10 mg/kg·BW simvastatin, and the rest 2 groups with equivoluminal bacterial-free water, 16 rats for each group, respectively. After 8 weeks, the levels of Leptin, MC4 R, MTTP, POMC, PPARγ DNA methylation were analyzed by bisulfite sequencing PCR(BSP) method, and the mRNA expression was detected by Q-PCR. Results showed as follows:(1) During the animal experiment, Cyclocarya paliurus(Batal.) Iljinskaja polysaccharide decreased the DNA methylation levels of LEPTIN, MC4 R, MTTP, POMC, PPARγ in liver(P<0.05 or P<0.01) and showed certain dose effect relationship;CPP showed no influence on these genes in adipose tissue except Leptin, and increased the DNA methylation level of POMC, when compared to the hyperlipidemic model group.(2) The results of Q-PCR showed that Cyclocarya paliurus(Batal.) Iljinskaja polysaccharide significantly restrained the mRNA expression of LEPTIN, MC4 R, MTTP, PPARγ(P<0.01 or P<0.05) in liver or adipose, and showed no influence on the POMC mRNA expression in both tissues.(3) The correlation analysis results showed that, in liver tissue, the DNA methylation levels of MC4 R, POMC, PPARγ showed negative correlation with mRNA expression, and the correlation is very weak, but the MTTP gene showed positive correlation; In adipose tissue, the DNA methylation levels of LEPTIN, MC4 R, POMC, PPARγ were positively related to the mRNA expression, LEPTIN, MC4 R had little correlation, and the correlation of POMC is extremely weak, However, PPARγ gene showed extremely significant correlation, and the MTTP gene showed a very weak negative correlation.(4) Cyclocarya paliurus(Batal.) Iljinskaja polysaccharide decreased the activities of DNMTs in adipose(P<0.05) and showed no regulation on liver in rats. The correlation analysis results showed that: the DNA methylation levels of LEPTIN、MC4R、PPARγ gene showed extremely significant correlation with the activities of DNMTs in both adipose and liver tissue(P<0.05), MTTP、POMC gene showed a weak correlation. |
