Objective:Established prostate cancer PC-3 cells vasculogenic mimicry model, in vitro,and to explore the mechanism of COX-2regulating prostate cancer PC-3 cell forming vasculogenic mimicry,provide a theoretical basis for the clinical application of anti tumor drugs.Method:1.Addition of celecoxib in prostate cancer PC-3 cells three-dimensional cultured model,through inverted microscope observe the effect of COX-2 inhibitor celecoxib act on prostate cancer PC-3 cells form vascular mimicry, proliferation inhibitory effect of celecoxib was detected by MTT technology,apoptosis effect of celecoxib on PC-3 cells apoptosis was detected by using flow cytometry and the effect of celecoxib on PC-3 cells could induce the expression of MMP-2 and Eph A2 which was detected by using Western Blot technology.2.We adding exogenous PGE2 on the foundation of the inhibitory action bringed by celecoxib,and observe the exogenous PGE2 impact on prostate cancer cell line PC-3 to form the vasculogenic mimicry, the effect on the proliferation, cell apoptosis,and the expression of Eph A2 and MMP-2.Result:1.The ability of forming vascular mimicry of PC-3 cells will weaken after adding COX-2 Inhibitor Celecoxib,and Celecoxib caninhibit the proliferation of PC-3 cells and the Celecoxib's effect of inhibit proliferation of PC-3 cells have a concentration and time dependent manners,meanwhile celecoxib can promote the apoptosis of PC-3 cells,and can make prostate cancer PC-3 cells to reduce the expression of COX-2?Eph A2 and MMP-2.2.After adding exogenous PGE2 on the premise of celecoxib's basic effect, the ability of PC-3 cell to form vasculogenic mimicry will partly recover which is observed by inverted microscope,can restore the inhibited proliferation by celecoxib, the apoptosis of PC-3 cell will significantly decrease and the number of S phase and G2/M phase cells increased significantly, and can restore the expression of Eph A2 and MMP-2 of PC-3 cells.Conclusion:1. In summary, inhibiting COX-2 can suppress the ability of prostate cancer PC-3 cell forming vasculogenic mimicry.2.The COX-2 participate in prostate cancer PC-3 cell line's growth and apoptosis.Further,it also participate in regulate the expression of Eph A2 and MMP-2.
