Objective: To investigate the expression and relationship of NLRP3 inflammasome-related Apoptosis-associated Speck-like protein containing a CARD(ASC), Cysteine aspartate-specific proteinase(Caspase-1), Interleukin-1?(IL-1?)with markers of myocardial injury including Troponin T(c Tn T), Creatine kinase-MB(CK-MB), Brain Natriuretic Peptide(BNP) in myocardial tissue and serum in rat model of acute myocardial infarction.Methods:To establish the acute myocardial infarction model of rat via the ligation of coronary artery of rat, which includes the control group,fake surgery group, and acute myocardial infarction(AMI) group. The blood at 0h, 12 h and 24 h hours after infarction were collected from abdominal aorta, the activity of myocardial injury markers including creatine kinase-MB, Troponin T, Brain Natriuretic Peptide(BNP), ASC,Caspase-1,and IL-1? in the serum were analyzed. The myocardial necrosis area at 24 hour after infarction was extracted for histopathological analysis including HE staining and TUNEL staining for apoptosis of myocardial cell. The expression of ASC, Caspase-1 and IL-1? in the myocardial necrosis area were analyzed by western blot. The expression of c Tn T?CK-MB and BNP in the myocardial necrosis area were analyzed by ELISA. The concentration of ASC, Caspase-1, IL-1? as well as myocardial injured markers including c Tn T, CK-MB, BNP in the serum of rat were examined by ELISA. The relationship of ASC,Caspase-1 and IL-1? with c Tn T, CK-MB, BNP in serum at 24 h after AMI were determed by SPSS.Results: Compared to the control group and fake surgery group,there are visible myocardial infarction tissues in AMI group. The histopathology results have shown the necrosis, edema and neutrophil granulocyte infiltration in the myocardial infarction area. The TUNEL staining has verified the apoptosis cells found in the myocardial ischemia tissue. Compared with the controlled group and fake surgery group, the expression of ASC, Caspase-1, IL-1?, c Tn T, CK-MB, and BNP were remarkably increased in the myocardial necrosis area of rat model(P<0.05). The concentrations of ASC, Caspase-1 and IL-1? in the serum were significantly increased with time-dependent manner at 12 h and 24 h after myocardial infarction. At 24 h after AMI, positive correlation between ASC and Caspase-1(r=0.711, P<0.01), ASC and IL-1?(r=0.871, P<0.01), Caspase-1 and IL-1?(r=0.796, P<0.01) were found in the serum. The concentration of ASC, Caspase-1 and IL-1? in serum were positively related to the myocardial injury markers including c Tn T(ASC vs c Tn T,r=0.957,P<0.01;Caspase-1 vs c Tn T,r=0.953,P<0.01;IL-1? vs c Tn T,r=0.953,P<0.01), CK-MB(ASC vs CK-MB,r=0.938,P<0.01;Caspase-1 vs CK-MB,r=0.934,P<0.01;IL-1? vs CK-MB,r=0.962,P<0.01)and BNP(ASC vs BNP,r=0.929,P<0.01;Caspase-1 vs BNP,r=0.927,P<0.01;IL-1? vs BNP,r=0.969,P<0.01)at 24 h after AMI.Conclusion: The expressions of ASC, Caspase-1 and IL-1? were remarkably enhanced in acute myocardial infarction, which is positively related to the myocardial injury markers c Tn T, CK-MB and BNP. All these factors might jointly contribute to the occurrence and development of acute myocardial infarction.
