Objective: By observing the PD rats induced by rotenone behavior change and gastrointestinal dysfunction, measuring the relevance of gallbladder contraction element(CCK) in serum, the new fat factor(Nesfatin-1) content and TH mean optical density of the substantia nigra striatum to analysis whether it can be used as PD early diagnosis index;Through comparing TH average optical density value in different parts at the same time,we may explore the pathogenesis of early PD.Methods: Adopt the principle of random 84 rats were divided into experimental group and 42 of solvent group. The experimental group with the back of the neck subcutaneous injection concentration of 2mg/ml of rotenone sunflower oil emulsion, injection volume such as sunflower oil emulsion solvent group, after 0, 2 weeks, 4 weeks, 6 weeks collect the rat feces within 2 hours, respectively, calculate the percentage of waste water, at the same time the determination of rat gastric residue weight/body weight percentage, step test and suspension test to compare the experimental group and solvent group rats behavior change,using immunohistochemical method in different period of the rectum, test chest pulp and substantia nigra striatum tyrosine hydroxylase(TH) the average optical density value, Elisa method was used to detect ghrelin CCK and Nesfatin-1 content.Pearson analysis method was used to analyze the relations between the CCK? Nesfatin-1 and tyrosine hydroxylase in the brain.Results: Behavioral testing found that the experimental group rats weight decreased significantly(P < 0.05);The experimental group hanging score was significantly decreased(P < 0.05), the most obvious was especially in 2 ~ 4 weeks,and dropped from 4 ~ 6 weeks, the experimental group rats stride number was decreased significantly(P < 0.05), and solvent group rats weight, hanging and step experiment had no obvious change(P > 0.05).The testing of Waste water and weight percentage of gastric contents found that from the 2 weeks the waste water content of the experimental group was significantly decreased(P < 0.05) and the percentage of gastric contents weight significantly increased(P < 0.05).Using ELISA method,we found that compared with the solvent group, the serum contents of CCK and Nesfatin-1 in experimental group rats became significantly higher(P < 0.05) from the 2 week, but the CCK in the 6 week comparing with the 2 week group, but the content of Nesfatin-1 was gradually declined;solvent group were no significant change(P > 0.05).Immunohistochemical results from the study showed that the TH average optical density value of the rectum, breast pulp and substantia nigra striatum was significantly decreased(P < 0.05), and especially in rectum showed a significant decline from the 2 week;and in solvent group,there was no significant change(P > 0.05).From the dynamic changes of the serum CCK content of experimental rats and the average optical density value of TH,we found over time it showed a negative correlation r = 0.953,(P < 0.05).There was no obvious correlation between the Nesfatin-1 content in the experimental group and the average optical density value of TH r = 0.530,(P > 0.05).Conclusion: It is increased significantly of CCK content in the early Parkinson's disease rats serum, and the content of CCK in the serum is negative correlation with dopaminergic neurons in the brain, hinting CCK in serum can be used as a possible predictor of early PD.
