| Objectives: To explore the application of curcumin and cisplatin plus on A549 lung cancer cell invasion and metastasis, and to explore the influence of the joint application of the two drugs MMP- 9, the influence of E- cadherin protein.Methods:MTT assay was performed to detect the effects of cur cumin, cisplatin alone and the two combination on A549 lung canc er cell proliferation; Transwell assay was performed to detect the ef fects of curcumin, cisplatin alone and the two combination on the i nvasion and metastasis of lun-g cancer cells; Western blotting was used to detect the protein expression of MMP- 9,E-cadherin?Results: 1. Curcumin, cisplatin alone and two medicine combined application of A549 lung cancer cell proliferation effect: 5?mol/L, 10?mol/L,20?mol/L,40?mol/L of curcumin on A549 lung cancer cell proliferation inhibition rate was respectively 6.50%?1.06%,11.70%? 0.88%,22.97%?0.82,27.93%?0.94%;The concentration of four different curcumin group compared with control group,(p<0.01);The difference was statistically significant;Four kinds of different concentrations of curcumin compared with between groups,(p<0.05),the difference was statistically significant;1mg/L,2mg/L,4mg/L of cisplatin on A549 lung c a n c e r c e l l p r o l i f e r a t i o n i n h i b i t i o n r a t e w e r e r e s p e c t i v e l y 7.12%?0.86%,20.07%?1.14%,26.88%?0.51;Three kinds of different concentrations of cisplatin group compared with control group,(p<0.01),the difference was statistically significant;Three different concentrations compared between cisplatin group,(p<0.01),the difference was statistically significant;Curcumin(20?mol/L) combined cisplatin(respectively 1mg/L?2mg/L?4mg/L)in A549 lung cancer cell proliferation inhibition rate respectively was 28.37%?0.57%,39.72%? 0.64%,46.27%?0.86%,compared with combination group and the control group,(p<0.01),the difference was statistically significant;Compared with single drug group, combination group(p<0.01),the difference was statistically significant;2. Curcumin, cisplatin and the two combination on A549 lung cancer cell invasion and metastasis of impact:curcumin group(20?mol/L), cisplatin group(2 mg/L)and joint(the concentration of curcumin is 20?mol/L+the concentration of cisplatin is 2 mg/L) attacks inhibition rate of A549 lung cancer cell were respectively 38.62%±0.23%,36.52%± 0.33%,63.78%±0.59%,compared with single drug group, combination group(p<0.01),the difference was statistically significant;3.The influence of curcumin,cisplatin and the two combination on A549 lung cancer cell of MMP-9, E-cadherin protein: the protein grey value of the ratio of control groups ?the curcumin group(20?mol/L)?cisplatin group(2 mg/L)and joint(the concentration of curcumin is 20?mol/L+the concentration of cisplatin is 2 mg/L)were respectively:MMP-9:0.768?0.047,0.654?0.104,0.684?0.008,0.444?0.104,E-cadherin: 0.603?0.170,0.792?0.050,0.784?0.045,0.879?0.110; Compared with single drug group, combination group(p<0.01),the difference was statistically significant.Conclusions: Curcumin,cisplatin and the two combination can inhibit the proliferation of A549 lung cancer cell, and within a certain concentration, the higher the effect of concentration, the stronger the proliferation inhibition of lung cancer cells, joint application effect is better than alone;Curcumin,cisplatin and the two combination can inhibit invasion and metastasis on lung cancer cells, but the combined was superior to the alone.Curcumin and cisplatin can make the cut, MMP-9 protein that E-cadherin rise, and after the combination effect is superior to use.Curcumin,cisplatin and the two combination can inhibit the invasion and metastasis of A549 lung cancer cell.its mechanism may be related to make MMP-9 protein decreased,and E-cadherin protein increased. |
