Objective This study is aimed to investigate the effect and mechanism of ellagic acid on the proliferation, migration and invasion of human pancreatic carcinoma PANC-1 cells, and to investigate the effect of ellagic acid on the pancreatic cancer PANC-1 xenograft of athymic nude mice.Methods (1) Investigate the effect of ellagic acid on the activity of human pancreatic carcinoma PANC-1 cells by CCK-8 assay. (2) Investigate the effect of ellagic acid on the repair ability of human pancreatic carcinoma PANC-1 cells by Wound healing trial. (3)Investigate the effect of ellagic acid on the migration and invasion of human pancreatic carcinoma PANC-1 cells by Transwell assay. (4) Investigate the effect of ellagic acid on the expression of E-cadherin and Vimentin protein in PANC-1 cells by Western blot assay. (5) Tumor xenografts of nude mice were established by subcutaneous inoculation of pancreatic cancer PANC-1 cells in the logarithmic phase into the Back of the nude mice near the hind limb.2 weeks after inoculation, Mice were randomized to one of four treatment groups with different concentration of ellagic acid treated. Measure the long diameter and short diameter of the subcutaneous transplantation tumors with vernier caliper, then calculate the tumor volumes. And draw the growth curve, and the record survival time.Results (1) With the increase of the concentration of EA,, the activity inhibition rate is higher, appearing a dose-dependent manner with statistically significant difference (P<0.05). (2) The wound healing of the control group was far wider than that in the treated group. With the decrease of EA concentration, the wound healing is wider. (3)With the increase of the concentration of EA, the migration inhibition rate is higher (P<0.05). With the increase of the concentration of EA and the extension of time, the invasion inhibition rate is higher(P<0.05). (4) Compared with the control group, the expression of Vimentin in PANC-1 cells was inhibited, and the expression of E-cadherin protein was enhanced in the treated group. (5)Ellagic acid significantly inhibited the growth of the tumor volumes:with the increase of EA concentration and the extension of time, the inhibition of the growth of the tumor volumes is higher (P<0.05). Ellagic acid significantly extended survival time of the pancreatic cancer PANC-1 xenograft of athymic nude mice (P<0.05).Conclusion Ellagic acid significantly inhibited the activity of the PANC-1 cells in a dose-dependent manner in vitro. Ellagic acid significantly inhibited the repair ability of the PANC-1 cells in vitro. Ellagic acid significantly inhibited the migration of the PANC-1 cells in a dose-dependent manner in vitro.Ellagic acid significantly inhibited the invasion of the PANC-1 cells in a dose- and time-dependent manner in vitro. Ellagic acid can the enhance expression of E-cadherin protein and inhibit the expression of Vimentin in PANC-1 cells. The potential mechanism might to be related to arresting the EMT process on PANC-1 cells, Ellagic acid significantly inhibited the growth of the pancreatic cancer PANC-1 xenografted tumor in a dose- dependent manner.
