| Objective: To observe the levels of thymic stromal lymphopoietin(TSLP), Th2 type cytokine interleukin-4(IL-4), Th1 type cytokines tumor necrosis factor g(TNF- g) in peripheral blood and total serum Ig E of patients with allergic rhinitis(AR) experimentally, analyze the correlations of TSLP with IL-4, TNF-g and total serum Ig E, and discuss the clinical significances of TSLP and the pathogenesis of AR.Methods: The specimens for this experimental research came from patients in the department of otolaryngology in Yueyang City Second People’s Hospital of University of South China. After defining strict diagnostic criteria, inclusion criteria and exclusion criteria, 30 cases with AR(20 males and 18 females, with a mean age of(34.51±13.71)) and healthy controls(11 males and 19 females, with a mean age of(30.71±8.52)) were collected. Enzyme-linked immunosorbent assay(ELISA) was employed to detect the levels of serum TSLP, IL-4, TNF-gand serum total Ig E of 30 cases with AR and 30 healthy controls and perform correlation analysis.Results: 1.The relative expression level of TSLP in peripheral blood of the experimental group was 27.5±2.49 and 18.86±2.01 in control group,The expression level of TSLP was significantly higer in experimental group(P<0.01);2.The relative expression level of IL-4 in peripheral blood of the experimental group was 44.65±5.24 and11.62±1.99 in control group,The expression level of IL-4 was significantly higer in experimental group(P<0.01);3.The relative expression level of TNF-g in peripheral blood of the experimental group was 30.89±5.85 and 86.12±6.88 in control group,The expression level of TNF- g was significantly higer in control group group(P<0.01);4.The relative expression level of serum total Ig E in peripheral blood of the experimental group was 47.14±5.32 and 18.78±1.35 in control group,The expression level of serum total Ig E was significantly higer in experimental group(P<0.01);5.Compared with the control group, the levels of TSLP, IL-4 and serum total Ig E of the experimental group were significantly increased, while that of TNF-g was greatly decreased,showingstatistically significant differences(P<0.01). 5.Compared with the control group, TSLP was positively correlated with the levels of IL-4of(r=0.688, P<0.01), while TLSP was negatively correlated with the level of TNF-g(r=-0.725, P<0.01),and IL-4 was negatively correlated with the level of TNF- g(r=-0.656, P<0.01), while IL-4 positively correlated with the levels of serum total Ig E(r=0.384,P<0.05). But TSLP were not correlated with the levels of serum total Ig E(r=0.262,P<0.01).Conclusions: 1.the levels of TSLP in peripheral blood of Allergic rhinitis patients group were significantly increased,it is speculated that TSLP may be involved in the pathogenesis of AR.Its effect may be related by mutual influence and interaction among the secretion of Th cell subsets cytokines leads to immune imbalance;2.The detectionl of TSLP levels in peripheral, has positive significance in clinical diagnosis of Allergic Rhinitis(AR). |
