Purpose By detecting the relative expression level of blood plasma miRNA-126 in diabetic retinopathy(DR) patients, to analysis the relationship between miR-126 and the occurrence and progress of DR, to find the correlation on clinical data such as sex, age, the stage of disease and the process of disease, to discuss the estimated value of blood plasma miR-126's expression level in disease severity and prognosis.Method We collected 12 healthy adults as control group and 55 cases of patients suffered from diabetic only as experimental group including none-obvious retinopathy group about 13 cases, low-grade NPDR group about10 cases, moderate-grade NPDR group about 11 cases, high-grade retinopathy group about 12 cases and PDR group 9 cases. After extracting miRNA from blood samples, we examined miRNA-126 by the real-time PCR(RT-PCR)method. All the results were analysised by SPSS 19.0; a single-factor analysis of variance procedure was used to compare the miR-126 expression levels in different stage of DR; Pearson correlation analysis procedure was used to calculate the relevance between miR-126 expression level and clinical matters such as sex, age, disease stage and progress.Results MiR-126 expression level in all diabetic retinopathy groups were significantly lower than the healthy group(P<0.00). Compared with none-obvious retinopathy group, the 3 NPDR groups and PDR group miRNA-126 expression were all significantly lower(P<0.05). There is no remarkable difference among the NPDR groups and compared with it, the PDR group miRNA-126 expression is apparently descended(P<0.05). The Pearson relevance analysis indicated a notable correlation between plasma miRNA-126 expression and disease stage(r=-0.833, P<0.000) or progress(r=0.396, P=0.020), also uncorrelated factors such as sex, age(P>0.05)and so on. There was no age or sex difference between all groups about miRNA-126 expression.Conclusion Blood plasma miRNA-126 expression is decreased gradually accompanying to disease stage elevated and to disease history prolonged. Its expression level is irrelevant to sex or age. As miRNA-126 functions to maintain angiogenesis working normally and to inhibit inflammation, so it is supposed to effect the neovascularization and inflammation reactions in diabetic retinopathy. And miRNA-126 might be a new target spot or a biological marker for clinical diagnosis, severity, or prognostic assessment in diabetic retinopathy.
