Nanomaterials technology have the rapid development in recent years, their application fields and even extended to medical, textile, information technology and energy, etc. Nano materials, especially for its extraordinary properties gaining increasing attention on the application and basic research in the areas of medical imaging, disease diagnosis, drug delivery and cancer treatment. But before the clinical application of the nanometer materials, the possible biological toxicity of the tissue should have a systematized research to avoid harmful effect to human body health than it brings to the benefit of human society. Thus it is necessary to have a research of safety evaluation, quality control and toxicology for the material. At present, this study is focused on the detection of biochemical indexes and histopathology section, but there is no study on the toxicity of endogenous substances.The main achievements of this paper are as follows:1. This experiment research object is Fe3O4 nanoparticles which are widely used in biology and medicine. The male Wistar rats were exposure d by intraperitoneal injection, the accumulation of liver surface after exposure of rats with nanoparticles was found. And in a considerable period of time it cannot be ruled out in vitro. The biochemical indexes of serum were detected, It was found that the content of TG in the model group was significantly higher than that of control group. The total cholesterol did not change significantly in the early stage, and then the content decreased with the increase of exposure time. Methanol/chloroform solvent was used for the target lipid extraction, UPLC was used for separation, QTof was used for detection, the analysis of lipid was finished in 10 minutes. After the Pretreatment of the raw data, the analysis of PCA and PLS-DA was conducted. According to the results obtained by PCA and PLS-DA, the obvious differences between the two groups were found. The abnormal metabolite included six phosphatidyl choline, four lysolecithin and a sphingomyelin was found, and it proofed that Fe3O4 nanoparticles have an effect on lipidomics.2. A method for rapid determination of eight organophosphorus flame retardants in polypropylene (PP) plastics by UPLC-MS was established. Response surface methodology was applied as the experimental design strategy to screen and optimize the extraction variables which affected the extraction efficiency, such as the volume of the extraction solvent, the time for extraction, and the temperature of extraction.
