Objective:The aim of the study is to investigate the expression of gene m RNA in Acute myeloid leukemia HL-60 cell by using Arsenic trioxide and provide a new way to study the mechanism and treatment of leukemia.Methods:Effect of CCK-8 to detect different concentrations of arsenic trioxide on HL-60 cell proliferation inhibition; Using inverted microscope to observe the cell Real-Time PCR detection before and after treatment of MAD2/CDC20 m RNA expression in HL-60 cells.Results:Arsenic trioxide on the proliferation of HL-60 cell have significantly inhibited, and the inhibition rate is proportional to the dose. By using different dose of Arsenic trioxide,the expression of MAD2/CDC20 m RNA decreased.Compared with the control group,the expression of MAD2/CDC20 m RNA was statistically significant(P<0.05),so as the experimental groups(P<0.05).Conclusion:Arsenic trioxide can inhibit the proliferation of HL-60 cell, so that the expression of MAD2/CDC20 m RNA decreased.
