This paper mainly study on the quality analysis of different Forsythiae Fructus from Henan Province by Near Infrared Spectra Technique ? HPLC fingerprint technology and pharmacological experiments.The main content is divided into the following four parts:The first part is optimization of the origin prosessing methods of Forsythiae Fructus.Compared three kinds of different Forsythiae Fructus by comprehensive evaluation indexes of forsythin and forsythoside A,the result show that the raw-drying Forsythiae Fructu's content of forsythin and forsythoside A are both lower than the other two kinds of products.Meanwhile,Optimized origin processing methods of steam drying products and boil-drying products by comprehensive evaluation indexes of forsythin and forsythoside A.The result shows that optimum processing method for Steam drying products is steaming 25 min;and optimum processing method for boiled drying products is 5.6 times of water,boiling 14 min.In addition,considering Chinese herbal medicine processing enterprise often stoving herbal medicine through mass production process,so we combined the effect of different drying temperature on indicative ingredients of forsythia fructus with the actual situation,finally choose the optimum drying temperature is 80??The second part is the application of near Infrared Spectroscopy in the quality analysis of Forsythia Fructus.Established qualitative and quantitative analysis model of Forsythia Fructus by near Infrared Spectroscopy,model validation results are good.Discriminant analysis is applied to identify different Forsythia Fructus,the model can identify Forsythia Fructus of different processing methods clearly,the analysis result is accurate,the error rate is 0.Partial least squares is applied to establish the quantitative calibration model of forsythin and forsythoside A.The correlation coefficients(R2)are 0.94825 ? 0.99489,the root mean aquare error of cross validation(RMSECV)are 0.02152 ? 0.26895,the root mean aquare error of calibration(RMSEC)are 0.00991 ? 0.182,the root mean aquare error of prediction(RMSEP)are 0.00750?0.0898,the performance index(PI)are 97.0?96.4.The model can predict the content of the sampleaccurately.Near Infrared Spectroscopy is convenient?accurate and non-pollution,easy to implement online testing,so it's feasible to be used for quality analysis of Forsythia Fructus.The third part is the study of HPLC fingerprint of Forsythia Fructus.First,established HPLC fingerprint of different Forsythia Fructus,using "chromatographic fingerprint similarity evaluation software(2004A Edition)" to calculate the similarity between HPLC fingerprint of different Forsythia Fructus and its common patterns.The result shows that different Forsythia Fructus have similar chemical compositions,but the contents differ significantly.Sun-dried products are relatively low similarity,Steam drying products and boiled drying products are ralatively high similarity,but all similarity are more than 0.9,this indicate It that the chemical composition of the samples are in good consistency,the quality more stable.Second,established HPLC fingerprint of Forsythia Fructus in different polar fractions,using "chromatographic fingerprint similarity evaluation software(2004A Edition)" to analysis the fingerprint,the similarity of each polar components is relatively high,chemical compositions between different polar fractions is significant difference.This experimental method is accurate,reliable and repeatable,the built HPLC fingerprint can fully reflect the distribution of the chemical composition of Forsythia Fructus,and it is suitable to extend to more quality analyze studies of chinese medicine.The fourth part is the study of correlation between HPLC fingerprints and Antibacterial pharmacological effects of Forsythia Fructus in different polar fractions.Experiments using Escherichia coli bacteria as the experimental bacteria to compare the antimicrobial effect of Forsythia Fructus in different polar fractions.Combined HPLC fingerprint with antimicrobial effect,using principal component analysis?cluster analysis and other statistical methods to speculate major chromatographic peaks correlated with antibacterial effect.
