Objectives Thyroid carcinoma is the most common malignant tumor of the head and neck, and the incidence increased year by year. A variety of factors such as pathological classification, different clinical stage and presence of lymph node metastasis are closely related to the development and prognosis of thyroid cancer. This experiment mainly discusses the expression of Periostin, Slug and E-cadherin in thyroid papillary carcinoma,and analyzes the relationship with clinical pathological characteristics, clinical treatment and prognosis of thyroid carcinoma judgment for providing certain theoretical support.Methods I collected 144 cases of thyroid papillary carcinoma tissue samples by after the operation diagnosed by experienced pathologist from the affiliated hospital of North China University of Technology in February 2009- February 2015, which including male patients 35 cases, female patients 109 cases, aged 21 to 81, and the average age of 45.7years old. 66 cases without lymph node metastasis, and lymph node metastasis occurred in78 cases. In accordance with international standards of cancer alliance(UICC) TNM staging, 62 cases?period, 23 cases?period, 58 cases ? period, and ? period in 1 case.Patients were not accepted by chemical treatment. At the same time set up nodular goiter(70 cases), thyroid adenoma(70 cases) and normal thyriod tissue(100 cases) as control group. By the immunohistochemical staining SP method detecting the expressions of Periostin, Slug and E-cadherin in the experimental group and the control group. The result of the experiment data using SPSS 13.0 statistical software for processing analysis.Results 1 The positive expression of Periostin protein is yellow or brown, and locating mainly in the cell nucleus. The expression, Periostin protein, in papillary thyroid carcinoma(83.8%), nodular goiter(10.0%), thyroid adenoma(0) and normal thyroid tissue(0). There is significant difference(P<0.05). The expression of Periostin protein is correlated with TNM classification and Lymph-node metastasis. There is significant difference(P<0.05). And it irrelates to age, gender and tumor size. There is not significant difference(P>0.05). 2 The positive expression of Slug protein is yellow or brown, and locating mainly in the cell nucleus. The expression, Slug protein, in papillary thyroid carcinoma(79.2%), nodular goiter(0), thyroid adenoma(0.7%) and normal thyroid tissue(1.0%). There is significant difference(P<0.05). The expression of Periostin protein is correlated with TNM classification and Lymph-node metastasis. There is significant difference(P<0.05). And it irrelates to age, gender and tumor size. There is not significant difference(P>0.05). 3 The positive expression of E-cadherin protein is yellow or brown,and locating mainly in the cell membrane. The expression in papillary thyroid carcinoma of E-cadherin protein(11.1%), meanwhile that of nodular goiter, thyroid adenoma and normal thyroid tissue(100%) are the same. There is significant difference(P<0.05). 4 In the experimental group, the high expression of Periostin was positively correlated with high expression of Slug(rs=0.367, P<0.01); the expressions, Periostin and E-cadherin,were significantly negative correlation(rs=-0.672, P<0.01); the expressions, Slug and Ecadherin, were significantly negative correlation(rs=-0.472, P<0.01).Conclusions 1 Periostin may have played an important role in promoting tumor invasion and metastasis in thyroid papillary carcinoma, which can be used as candidate genes of cancer metastasis of thyroid papillary carcinoma; 2 Periostin participated in the transformation of epithelial mesenchymal in thyroid papillary carcinoma, and may through the Slug-E-cadherin-EMT way to play a role in thyroid papillary carcinoma.Therefore, detection of expression level of Periostin in thyroid papillary carcinoma tissue might be for clinical diagnosis and evaluating patients' clinical prognosis.
