Objectives Objective to establish the model of hypobaric hypoxia at the attitude of 7000 meter injury on Wistar,to characterize the effect of hypobaric hypoxia to EMT in epithelial tissue,whether to block EMT in epithelial tissue to decrease the invasion and metastasis of epithelial malignant tumors and the occurrence of tissue and organ fibrosis.Methods 1 Isolation of oral mucosa epithelial tissue of Wistar with hypobaric hypoxia use the method of Dispase ?digestion. 2 The expression of mi R-92 a detected by Quantitative Real-time PCR. 3 The genes expression levels of the HIF-1?,E-cadherin and Vimentin detected by reverse transcription polymerase chain reaction. 4 The proteins expression levels of the HIF-1?,E-cadherin and Vimentin detected by Western Blot. 5 The morphological changes of the oral mucosa epithelial tissue inspected by HE staining.Results 1 Dispase ? could digest oral mucosa epithelial tissue, and basal layer cell was not destroyed. 2 q PCR results:compared with control,the expression of mi R-92 a was decreased in hypoxia 12 h and 24 h,the difference was statistically significant(P<0.05). 3PCR results:compared with control,the expression of HIF-1? and Vimentin increased in hypoxia 12 h and 24 h,whereas E-cadherin was decreased,the difference was statistically significant(P<0.05). 4 Western Blot results:compared with control,the expression of HIF-1? and Vimentin increased in hypoxia 12 h and 24 h,whereas E-cadherin was decreased,the difference was statistically significant(P<0.05). 5 HE staining results:compared with control, the cell morphology of oral mucosa epithelial cells was not obviously abnormal,and there was no mitotic figure in basal layer and stratum.Conclusions Hypoxia injury induces HIF-1? accumulation, down regulates miR-92 a expression, and then promotes epithelial-mesenchymal transtion of epithelial tissue,to provide a new way to reduce the invasion and metastasis of epithelial malignant tumor and the occurrence tissue and organ fibrosis.
