| Objectives 4 kinds of Wistar rat animal model of rheumatoid arthritis were comparison order to choose a more ideal animal model, to provide experimental evidence for the application of animal models in the study of the mechanism of rheumatoid arthritis and the evaluation of therapeutic effect.Methods 100 five-week-old female Wistar rats were adaptive fed one week,and were randomly divided into five groups.The five groups were adjuvant arthritis model group(A group), collagen-induced arthritis model group(group B), ovalbumin-induced arthritis model group(group C), rheumatoid arthritis cold model group(group D)and the normal control group(E group).each group has 20 rats respectively, according to their respective methods of modeling, after modeling all the animals were fed under the same conditions.Which include feeding,eating and drinking freely ordinary feed timing quantitative feeding,and select the adequate light, well ventilated experimental environment to feeding.Records the rats general state everyday after the experiment,including hair neatly gloss, body weight, slow degree, mental status, ears, tail, feet whether joint swelling and other inflammatory response.Then respectively to arthritis score of each group of rats, measure the weight of rats in each group, joint swelling,spleen weight, and calculate the spleen cell proliferation rate and the spleen weight coefficient at the 7, 14, 21 and 28 days. Every rats’ right toe will done X-ray observations,HE staining was performed on synovial tissue of the joint. Meanwhile heart blood and serum was separated to detect the content of serum IL-10 and IL-17 with ELISA method.Results Comparison of the results within the group:After modeling animal appetite loss,slow, lack of energy, fur and dull,the weight decreased slightly. Rats in group A induced inflammation after about 7 days the right toe swelling reached the peak and gradually reduce for 3 days, then the right toe swelling again about 14 days, The side and front feet is swelling and progressive increased. The rats’ action was inconvenience and the fur lose luster. The skin ulcer in rats’ right toe occurs around 21 days.At 28 days, further pathological changes and ulcer surface to further expand.The 14 days rats’ spleen cell proliferation rate have significant difference compared with 7 days and 21 days(P<0.05).The rats’ body weight, right toes swelling degree and spleen weight coefficient in each stage has a significant difference in group B(P<0.05), animal sensitization after about7 days, some rats ankle skin redness, minor swelling. Most of the rats ankle swelling, foot skin shiny, hyperemia, restricted joint activities after about 14 days.The joint swelling continue to increase about 28 days. The rats’ body weight, right toes swelling degree,spleen weight coefficient at 7 days have significant difference compared with that at 14 days, 21 days and 28 days.The rats’ body weight, right toes swelling degree and IL-10 in each stage has a significant difference in group C(P<0.05). The swelling at 7 days and 14 days have significant difference compared with that at 21 days and 28 days.The IL-10 concentration at the 14 th day has significant difference compared with that at 7, 21 and 28 days.The rats’ body weight, arthritis scoring increment rate, spleen cells and IL-17 in each stage has a significant difference in group D(P<0.05).The arthritis score at 7 days have significant difference compared with that at 14 days and 21 days.At the seventh day the spleen cell proliferation rate had significant difference compared with 14 days and 28 days.The spleen cell proliferation rate began to increased on 7 days and after it began to decline on 14 days then about 28 days it is began to increase.The IL-17 concentration at the 14 th day has significant difference compared with that at7, 21 and 28 days.Comparison of the results between groups:At the seventh day five groups rats’ arthritis of rats score, the spleen cell proliferation rate and spleen weight coefficient have a significant difference between groups(P<0.05). Most rats of A group and D group ankle swelling, foot skin shiny, hyperemia, joint activity limitation at the fourteenth day. Five groups of rats’ right toe swelling, arthritis score, spleen cell proliferation rate, the concentration of IL-10 and IL-17 has a significant difference between the groups(P<0.05).At the twenty-first day rats’ joint swelling continue to increase, the rats of A group appear debaucjed of skin ulcer. Five groups of rats’ right toes swelling degree, arthritis scoring,spleen cell proliferation rate and concentration of IL-10 has a significant difference between groups(P<0.05). At the twenty-eighth day the rats’ joint swelling continues to increase, the rats in A group skin ulcer and aggravating. Through the analysis of variance,five groups rats’ arthritis scoring, the spleen cell proliferation rate, spleen weight coefficient, IL-10 and IL-17 concentration has a significant difference between groups(P<0.05).Conclusions AA model group, CIA model group, ovalbumin induced arthritis model and the model group of rheumatoid arthritis can be successfully set up in female Wistar rats.The rats’ joint swelling of AA model group is obvious. AA model performance is closer to human RA. Then the clinical pathology, the course of disease and immunological changes are similar to human RA.The modeling process is simple and stable, which is an ideal animal model of rheumatoid arthritis.The joint swelling of the model group was obvious in the rheumatoid arthritis model.Under the strict control of molding conditions, this model can also be used as the animal model of rheumatoid arthritis ideal.The rats’ performance of ovalbumin induced arthritis model is not very prominent, and the development course of short duration is not a good simulation of human RA, it cannot be comprehensive to provide experimental basis for rheumatoid arthritis occurring mechanism and treatment effect evaluation. |
