The clinical applications based on gene therapy have been studied to achieve the goal of treating or preventing some kinds of diseases. The successfully application of gene therapy is depended on the gene delivery vectors to a large extent. Liposomes, inorganic nanoparticles and cationic polymer nano-carriers have got extensive applications in gene therapy. This thesis is based on the cationic liposome and gold nanoparticles gene vectors, and the transfection efficiency of them is studied in vitro. The main points are outlined as follows:1. We added a certain proportion of cholesterol(Chol) into Dimethyldioctadecylammonium Bromide(DODAB) to synthesize gene vector. We demonstrated the stability of above liposome and its interaction with DNA by agarose gel electrophoresis assay. In vitro transfection experiments, it was found that the transfection efficiency of DODAB/Chol in HEK 293 cells and HeLa cells was not very low. Through MTT assay, it is demonstrated that the relative cell activity of HEK 293 cells after treated with DODAB/Chol.2. We synthesized the gold nanoparticles protected by cationic liposome DODAB/Chol by one step synthesis. We characterized the gold nanoparticles using UV-vis Spectrum, Transmission Electron Microscopy, Zetasizer nano ZS and interaction between gold nanoparticles and DNA by Agarose Gel Electrophoresis. In vitro transfection experiment verified that the efficiency of DODAB/Chol-AuNPs in HEK293 cells was greatly improved compared with that of DODAB/Chol. In vitro cell relative activity analysis results showed that DODAB/Chol-AuNPs did not induce higher cytotoxicity than that of DODAB/Chol.
