Objective To determine the expression levels of Eg5 in neuroblastoma and the effect of Eg5 specific inhibitor S-Trityl-L-Cysteine(STLC)on the cell proliferation and cell division in neuroblastoma. Methods Immunofluorescence and Western blot was performed to detect Eg5 expression in neuroblastoma tissues and cells. With the induction of STLC, cell morphology was observed by phase contrast microscope, cell proliferation, cell cycle and cell apoptosis were examined with CCK-8 assay and flow cytometry, MYCN expression was detected by FISH assay with STLC induction, nuclear staning by DAPI was performed to investigate the morphology change. Results Eg5 was expressed in neuroblastoma tissues and cells, with the induction of STLC, cell morphology will be changed and apoptotic bodies were observed.The Eg5 inhibitor effectively prevented the proliferation and arrests cells at G2/M phase,then promoted cell apoptosis, which were dose and time-dependent. STLC has no effect on the expression of MYCN. Conclusions Eg5 was expressed both in neuroblastoma tissues and cells, STLC suppressed the proliferation and cell division,arrests cells at mitosis and induced cell apoptosis. These data thus indicated Eg5 as a potential target for neuroblastoma treatment probably.
