Objective:To study the effects of triptolide on proliferation and apoptosis of Triple-negative Breast Cancer Cell Line MDA-MB-231.Methods: MDA-MB-231 cells were cultured in vitro and were treated for 48 h with 0,12.5, 25, and 50nmol/L triptolide, respectively. The percentage of cellular proliferation was detected with MTT; the percentage of cellular apoptosis was detected using TUNEL; the expression levels of Bax?Bcl-2 and p53 were detected using western blot.Results:Compared with the control,after treated with 12.5, 25, and 50nmol/L triptolide,the percentage of cellular proliferation was reduced from(100±0.00)% to(86.23±5.09)%,(50.24±2.90)%,(18.89±2.39)%, respectively; the percentage of apoptotic cells was increased from(3.40± 2.79)% to(10.12± 3.18)%,(53.61±9.01)%,(60.22±17.21), respectively; the expression levels of Bax and p53 protein were both up-regulated,while Bcl-2 protein was down-regulated,and they were all in a concentration-dependent manner.(each group compared with control group,p<0.05).Conclusion:TPL significantly inhibits the proliferation of MDA-MB-231 cells, and also enhances apoptosis of MDA-MB-231 cells.p53 protein, Bax protein and Bcl-2 protein all play a role in the apoptosis of breast cancer MDA-MB-231 cells treated with TPL.
