Objective: To detect the expression of GFAP in different times of temporal lobe epilepsy, to explore the possible role of HIF-1? in the astrogliosis induced by temporal lobe epilepsy.Methods:We used Li- Pilocarpine to induce chronic model of temporal lobe epilepsy.And the rat were randomly divided into two groups namely the experimental group and the inhibiting group which were applied with 2-MEM,the antagonist of HIF-1?. Animals were killed in different periods(3days?7days?14days?28days and 6 months) after SE to observeastrogliosis in hippocampus. To label astrocyte, GFAP immunohistochemical staining was applied. Then we compared the extent of astrogliosis in different periods of epileptogenesis and different groups through observing the results of staining with the light microscope.Results:1?After the temporal lobe epilepsy, the GFAP expression began to increase 3 days later,and about to 14 th day reached the peak,and then began to decrease gradually.The expression of GFAP in hippocampus still remains high 6 months later. 2?After the application of the antagonist of HIF-1?,the expression of GFAP in inhibiting group did not show significant change compared to the epilepsy group.Conelusions:1?The acute period is important for astrogliosis induced by epilepsy,and our experiment provide evidence for choosing the right time for intervention of astrogliosis induced by epilepsy.2?There is no significant connection between the up-regulation HIF-1? and astrogliosis induced by epilepsy.
