Background: MUC1 is a transmembrane glycoprotein expressed by the majority of mucosal epithelial cells and by hematopoietic cells.It is widely expressed in various epithelial tissues,such as respiratory tract, gastrointestinal tract,genitourinary tract and mammary glands.The features of protection, lubrication and immune regulation in epithelialium have been identified.Studies have indicated that MUC1 plays a role in airway infection and inflammation by diverse bacterium and viruses.The interaction between epithelial cells and dendritic cells,which leads to Th2 response, is of great importance in the pathogenesis of asthma.Cytokines like GM-CSF,TSLP are involved in the interaction.It is deserved to investigate whether MUC1 participates in the mechanism of chronic inflammatory airway diseases like asthma.Objective: To investigate whether IL-13 affects the secretion of GM-CSF by 16 HBE cell,and the role of MUC1 involved.Methods: 1. 16 HBE cells were stimulated by IL-13 with different concentrations(0,20,40,60ng/ml)for 24 h.Detect the supernatant of each group by ELISA. 2.Grouping design of MUC1 si RNA transfection experiment:(1) Control 16 HBEC,(2)Scrambled si RNA transfected 16 HBEC,(3)MUC1 si RNA transfectied 16 HBEC,(4)Control 16HBEC+IL-13,(5)Scrambled si RNA transfected 16HBEC+IL-13,(6)MUC1 si RNA transfected 16HBEC+IL-13.Then 16 HBE cells were stimulated by IL-13(20 ng/ml) for 24 h.MUC1 expression was detected by western bolt,while the secretion of GM-CSF was detected by ELISA.Results: 1. Compared with the control group,the secretion of GM-CSF decreaced after 16 HBE cells were stimulated by IL-13(p<0.05).However, there was no remarkable difference between each group with different concentrations of IL-13(20,40,60ng/ml. 2. After stimulated by IL-13 for 24 h,MUC1 protein expression of 16 HBE cells were reduced(p<0.05). 3. The secretion of GM-CSF decreased when 16 HBE cells were transfected by MUC1 si RNA(p<0.01).Conclusion: IL-13 could inhibit the secretion of GM-CSF in 16 HBE cells by decreasing the MUC1 protein expression.
