Objective: To observe the effects of EPO on myocardial cells and coronary microcirculation in rats with myocardial ischemia-reperfusion injury and its mechanism.Methods: Sixty male and female SD rats were randomly divided into false surgical group, IR model group and EPO treatment group. After MIR models were made,the EPO treatment group were given 50000U/kg rh EPO intraperitoneal injection while other groups were given equal physiological saline. Then take 5 rats from each group to have ink perfusion to make myocardial tissue biopsies to observe the myocardial capillary number ink perfusion. Other rats were sacrificed to take their myocardial tissue to analyze the contents of malondialdehyde(MDA) and superoxide dismutase(SOD); observe the pathological changes in the myocardial by HE staining; Western-blot measure the expression levels of bcl-2/bax protein; gauge the expression of bcl-2 and bax in myocardial tissue by immunohistochemistry.Results : 1)The comparison of EPO treatment group with IR model group showed that the pathological lesion of EPO treatment group was lighter;2)Compared to IR model group, the content of MDA decreased(P<0.05),while the expression of bax decreased not obviously; the content of SOD?expression of bcl-2 and bcl-2/bax decreased(P<0.05);3) Compared to IR model group, EPO treatment group`s number of capillaries ink perfusion in ischemia area obviously increased(P<0.05).Conclusion: EPO can protect MIRI rats myocardial cells. The mechanism may be related to its anti-oxidant function?increaseing expression of anti-apoptosis protein bcl-2 and improving coronary microcirculation.
