Objectives To explore the inside relation of Slow transit constipation( STC) and autophagy in Cajal interstitial cell.Methods 60 SD weighing200~205g ratswere randomlydivided into two groups, the control group and the treatment group. The treatment groupratsweregivenby gavage once a dayat a dose 50 mg/kg Berberine for 14 days according to 10 ml/kg of the weight of rats.The control group were given normal saline of the same quantity. To identity the effect of the STC rats, several methods were employed to test the STC pathological symptom, including the amount of faeces, the time of originfaeces dropping and the moisture content in the faeces.After the STC models established successfully, we dissected these rats of the two groups and the intestinal tract was used to explore the changes of Cajal interstitial cell by western blot and IHC to identify c-Kit expression. Finally, western blot and TEM were used to detect the autophagy flux in the Cajal interstitial cell.Statistical software SPSS 16.0were used to analyze the experimental data, data between two group was compared by the LSD- t test. The results are statistically differences in P < 0.05.Results 1 After 14 d treatment with physiological saline or Berberine by ig, the body weight between the two groups had no significant difference(P < 0.05).The data of three main point detection showed that the STC rats model were established successfully,with the delay of the first droppings time,the total amount droppings decreased in 6h after the treatment of black ink and the moisture content lost in the faeces of the STC rats.2 By IHC detection of the c-Kit in the ICCss,results showed that the ICCss in the STC group were changed with the morphological changes in per cell and decrease amount in the network structure. Results of Western blot also suggested that the expression of c-Kit,marker of ICCss,was downregulated in the STC group. It was for the first time to show that autophagy was observed in the ICCss when the Rats was in the situation of STC. 3After the TEM detection, we found that the autophagic flux and the amount of autophagosome was increased in the STC rats. Data of the positive autophagic ICCss and the number of autophagic ICCss in ICCss both increased obviously when compared with the control group. Results suggested that the autophagic cell death might relate to the causing of the STC rats.Conclusions 1Morphological changes of ICCss and the decreasing amount of ICCss network were observed in the STC rats. 2 In the rat model of slow transit constipation and intestinal cells of Cajal undergone significant autophagy phenomenon, which may be one of the causes leading to slow transit constipation caused by reducing the number of interstitial cells of Cajal. 3 slow transit constipation in rats free of Cajal interstitial cells increased Ca2 + ions, Ca2 + ions increase may be the cause of interstitial cells of Cajal autophagic death.
