Purpose : In our previous studies, deceased Annexin A1 expression was found in some oral squamous carcinoma cell lines when compared to the human immortalized oral epithelial cells(HIOEC). Annexin A1 has been reported to play a role in the inflammatory reaction by regulating cellular proliferation and apoptosis; furthermore, it has also been reported to play an important role in the development of variety kinds of cancers. Unfortunately, the role of Annexin A1 in oral squamous cell carcinoma(OSCC) is not fully understood.In this study, the expression of Annexin A1 was detected in some oral squamous cell carcinoma lines, further biological studies on OSCC lines were performed to interfere with the Annexin A1 expression in vitro and in vivo.Methods 1?Annexin A1 expression was detected in OSCC lines and HIOEC by Western blot and real-time PCR.2?Plasmid vectors containing si RNA sequence against Annexin A1 or Annexin A1 gene were built, and virus vectors were also built. OSCC lines were infected by the vectors to get the cells with Annexin A1 overexpression or si RNA.3?Cell growth was examined by MTT and real-time cell analysis. Xenograft tumors were also studied to investigate the cellular proliferation of OSCC cells interfered with si RNA against Annexin A1 in nude mice.Results 1?Decreased expression of Annexin A1 in the cell lines of HB96, HN4, HN13, HN30 and CAL27 was found when compared to the HIOECs.2?si RNA interference against Annexin A1 was used in the HB96 cell line to decrease the Annexin A1 expression level. Overexpression of Annexin A1 was used in the HN13 and CAL27 cell lines to increase the Annexin A1 expression level. The Annexin A1 expression level was verified by Western blot and real-time PCR.3?Increased abilities of cellular proliferation and migration were found in the HB96 cells when interfered by si RNA against Annexin A1. Decreased abilities of cellular proliferation, migration and xenograft tumors in nude mice were found in the CAL27 cells with Annexin A1 overexpression by gene transfection compared to their control.Conclusions 1?Decreased Annexin A1 expression is common in OSCC cell lines.2?Annexin A1 inhibits the cellular proliferation in OSCC cell lines, which acts like a tumor suppress gene. After overexpression of Annexin A1, the OSCC cells have a faster cellular proliferation, migration and xenograft tumor formation.
