Protein is one of the most basic compositions of life, which is the most basic composition of physiological structures and it also bears the responsibility of life's maintenance. Researches show that the proportion of protein in human body is up to 18%. After all, protein is the highest content of organic matter in the human body. Besides that, protein can combine with a variety of small molecules, ions and biologically active substances. It is particularly important to note that the human serum albumin can be combined with a lot of exogenous drug molecules. It is essential for serum albumin to transport drug molecules as carriers. The research on the interaction between drugs and serum albumin has important practical significance for the development of new drugs.The method of spectral analysis has the advantages of high sensitivity, fast speed, less sample,simple operation and so on. Therefore, in this paper, the research on the subject of drug-protein interaction is carried out by fluorescence spectrum and ultraviolet absorption spectrum:(1) Based on the fluorescence spectra, the specific binding characteristics of the mutual role are studied and the types of the fluorescence quenching of the drug molecules with bovine serum albumin are also analyzed. After getting the fluorescence quenching curves of different drugs, the interaction between molecules was calculated. According to the non-radiative energy transfer theory, the distance between the molecules is calculated.(2) The method of fluorescence anisotropy is used in the interaction between fluorescent drug molecules and bovine serum albumin. In the interaction of the host-guest molecules, the binding constant and binding sites of the molecules were calculated from the view of guest. The spectral overlap in the fluorescence quenching method was effectively solved.(3) The effects on the conformation of serum albumin are studied by UV absorption spectra,resonance scattering spectra, Synchronous fluorescence spectra and three-dimensional fluorescence spectra.
