Objective: In recent years, the association between tumor associated neutrophils(TAN) and tumor occurrence, progress become a research hotspot, we found that neutrophil in tumor progression has a dual role. On the one hand, activated neutrophils can play a role in inhibiting tumor in many ways, on the other hand, neutrophils can also promote tumor formation, progression and metastasis. NLR was found to be correlated with tumor stage, pathological grade, histological type, and can be used as an independent predictor of prognosis. Neutrophil gelatinase-associated lipocalin(NGAL) has been confirmed play an important role in the initial stage of tumor cells, and it can be composed with matrix metalloproteinase-9 to promote tumor invasion and metastasis process. Therefore, the study aims to analyze NLR and the expression of TAN, NGAL in bladder tumor tissues, and the relationship between them and the clinical and pathological issues. Methods: To collected the specimen of bladder urothelial carcinoma, tissue sections and immunohistochemical staining. To research NLR and the expression of TAN, NGAL in bladder urothelial carcinoma, and analyze the relationship between them and bladder cancer stage, pathological grade and postoperative recurrence, gender, age, tumor number, incidence, Ki-67, smoking history and other related clinical and pathological issues. In this study, we used chi square test and Fisher's exact test to analyze the data, the difference was statistically significant with P<0.05.Result: 1. Immunohistochemistry showed that TAN and NGAL were positive in all 73 bladder cancer tissues. 2. By statistical analysis, the relationship between them and clinical stage, pathological grade, postoperative recurrence, Ki-67, smoking history has significant correlation(P < 0.05), and has no significant correlation with patient gender, age, number of tumors, incidence(P > 0.05). In the study, we found that the levels are different in different clinical stage and pathological grade, the higher clinical stage is corresponding with higher NLR and expression of TAN and NGAL.The levels of TAN, NLR, NGAL in patients with no recurrence of bladder cancer tissue are lower. The expression of TAN, NGAL is lower when the level of Ki-67 is low. Similarly, the levels of TAN and NLR was higher in bladder cancer tissues of patients with smoking history. Conclusion: 1. The expression of TAN, NGAL in bladder cancer tissues has different levels. 2. The levels of TAN, NLR, NGAL has significant correlation with clinical stage and pathological grade, postoperative recurrence, Ki-67, smoking history.they are expected to become biomarkers in terms of diagnosis, treatment and prognosis in bladder cancer. We will conduct large sample study to get more accurate boundary value, and to make them become the effective indexes of the diagnosis, treatment and prognosis in bladder cancer. 3. To further research the mechanisms of TAN, NGAL in bladder cancer formation, progress, invasion and prognosis, and provide theoretical basis for biological immunotherapy to make the treatment of tumor more effectively.
