Objective This study was to investigate SOCS1 in cervical cancer, precancerous lesion, cervical cancer cell lines and its clinical significance. Transfected recombinant plasmid of SOCS1 into cervical cancer cell line to observe its expression. To study the regulation of SOCS1 in the proliferation and metastasis of cervical cancer cell line, which could provide the basic theory for gene therapy of cervical cancer.Methods Chose the patients from Gynaecology Department of The Second Hospital of Tianjin Medical Univercity. Including 36 samples of low grade squmous intraepithelial lesions(LSIL), 30 samples high grade squmous intraepithelial lesions(HSIL), and 57 samples Cervical cancer(CC), dignosised by colposcopy protractor biopsy. The control group was normal cervical. These patients accepted uterus cut operation because benign uterus lesions. RT-PCR and western blot were used to analyse the expression level of SOCS1 in cervical cancer, cervical cancer lesion and the cervical cancer cell lines. Transfect recombinant plasmid of SOCS1 into cervical cancer cell lines. To observe the affection of SOCS1 recombinant plasmid on proliferation and metastasis of cervical cancer cell by MTT and cell scratch experiment..Results The first part: SOCS1 expression in cervical lesions and its clinical significance. SOCS1 expression of m RNA and protein in cervical carcinoma was significantly decreased compared with those expressions in the normal cervical tissue, low-grade intraepithelial neoplasia and high grade intraepithelial neoplasia(P < 0.05). The high grade intraepithelial neoplasia tissues was significantly decreased compared with those expressions in low grade intraepithelial neoplasia and normal cervical,(P<0.05).While there was no significant difference between low grade intraepithelial neoplasia tissue and normal cervical tissue(P > 0.05). The second part: The effect of SOCS1 on the biological behavior of Hela cell line. study the the proliferation and metastasis of cervical cancer cell with plasmid SOCS1.(1) Tranfect PC DNA3.1(+) SOCS1 into Hela cervical cancer cells, successfully achived the high expression of SOCS1, which had significant differences compared with those witout transfection and transfected with blank PC DNA3.1(+).(2) E7 m RNA and protein expression in Hela cell with SOCS1 plasmid was decreased significantly than control group.(3) There was no significant difference among witout transfection group, blank plasmid group and transfected with SOCS1 plasmid group in 24 hours while after 48 hours the multiplication rate and the transfer ability were dropped significantly in PC DNA3.1(+) SOCS1 groups.Conclusions(1) The findings suggest the different expression level of SOCS1 could closely associated with the occurrence of cervical cancer.(2) SOCS1 has an important role in inhibition of cancer cell proliferation, invasion through plasmid in Hela cells..(3) SOCS1 may plays an important role in the development, invasion and metastasis of cervical cancer, which could be used as cervical cancer gene therapy targets. Our reaearch established the basic theory for the futher research of gene therapy and assessment the extent of cervical cancer progression..
