ObjectiveBased on the basis of proteomics protein expresseddifferences betweenHeat-toxin Stagnation Syndrome AIDS patients and healthy people by iTRAQ technology, to observed characteristic changes in protein expression. to explore the efficacy of drugs and syndromes characterized by the expression of proteins between changes.Proteins were molecular markers with diagnostic and therapeutic significance for the evaluation of Heat-toxin Stagnation Syndrome AIDS and provide experimental evidence.MethodsIn this study, The AIDS patients of Heat-toxin Stagnation Syndrome are treated with Ai rehabilitation capsule combined with highly active antiretroviral therapy (HAART) for 48 weeks and to detect respectively (0 weeks-.48 weeks) clinical baseline indicators of the serum samples of whole blood and extraction such as WBC, HBV DNA, CD4+T, liver and kidney function. Based on the preliminary study of iTRAQ technique, To detect respectively differences in protein of AIDS Heat-toxin Stagnation Syndrome patients and healthy controls blood samples by means of using Western Blot techniques, to derivethe relative gray value bydoing comparison, the gray value on based on of ImageJ analysis using software gray value, Then to getmean+/-SD charts by way of entering therelative gray value relative gray value of the normal group and patient group into GraphPad Prism 5 software count or measurement data are analysised by SPSS 17.0 statistical software.Results1.Comparison of AIDS medication before Heat-toxin Stagnation Syndrome and normal control group, C5 in Heat-toxin Stagnation Syndrome of AIDS downregulated, and the difference was statistically significant (P<0.05), consistent with iTRAQ screening results; keratin 10 of AIDS downregulated of Heat-toxin Stagnation Syndrome, the difference was statistically significant (P<0.05), consistent with iTRAQ filter results.2.Heat-toxin Stagnation Syndrome of AIDS drugs in patients 48 weeks after the intervention in comparison with before treatment, CD4+T, CD 8+T stable increase, CD4+T statistically significant (P<0.05), CD8+T not statistically significant.3.The front and rear of Heat-toxin Stagnation Syndrome AIDS medication comparison, C5 in the Heat-toxin Stagnation Syndrome of AIDS medication upregulated, the difference was statistically significant (P<0.05).Conclusions1.The present study results AIDS Heat-toxin Stagnation Syndrome characteri zed by the presence of protein. Which are respectively C5? keratin 10, Both of w hich may be Heat-toxin Stagnation Syndrome biological.2.The present study results C5 protein expression levels and Heat-toxin Sta gnation Syndrome of traditional Chinese medicine diagnosis and treatment of AI DS treatment effect was consistent trend for Heat-toxin Stagnation Syndrome AI DS treatment has efficacy evaluation significance.3.The study of AIDS of Heat-toxin Stagnation Syndrome to verify the effi cacy of the intervention and the characteristic protein. The establishment of AI DS syndrome diagnosis model, improve and standardize the AIDS syndrome d ifferentiation methods, clinical syndromes of TCM AIDS diagnosis and evaluati on made outstanding contributions.
