As we all know,during the process of the Vcholerae invade the organization,it always needs to penetrate the mucin at first,then arrive the intestinal epithelial cells.However,Vibrio cholerae's flagella will break actively in the process of entering the mucus layer,We introduced the way of random transposon insertion,screen the mutant which could promote the motility in the 20%semisolid LB medium with 0.4%mucin,we screened the gene related to the synthesis of VPS,which may be used to promote the motility in mucin.In-frame deletions of the vpsA we completely construct the strains which cannot produce VPS.For the vpsR is one of the major regulators for VPS expression,we obtained the VPS overexpressed strains through overexpress vpsR.In the physiologic ways,we mainly focus on comparing the strains of?vpsA(negative),WT::Ptac-vpsR(positive)and WT's motility in the 20%LB semisolid medium with 0.4%mucin or not.Meantime,we also observe the three strains' activity from microcosmic aspect through Live Cell Imaging System.We found that mucin could promote Vibrio cholerae's motility,and VPS is the obstacle of Vcholerae move on the mucin.From the mouse model,we found that the VPS's expression in vivo also could influence V.cholerae's motility in mucin.Through RT-PCR,we analyze the expression of vpsA,vpsR,vpsT in the medium with mucin or not separately.Meantime record their growth curve in the medium with mucin or not,and found that Vcholerae might close the VPS's expression to influence its motility in mucin.At last,based on the chemotaxis,growth curve,different nitrogen and carbon resource,different surfactant and different genes,we obtain a tentative research around the mechanism of the mucin how to influence the V.cholerae's motility.And we considered that the V.cholerae could be chemotaxis by mucin during the process of Vcholerae move on the mucin so that promote its motility.Meanwhile,the mucin's rich nutrition also could promote the growth of strains.The viscidity and ability to combine free water of mucin could promote the Vcholerae's motility to an extant.The genes like responsible for the flagella,or motor protein,or chemotaxis,most of them influence the Vcholerae's motility in mucin through affect their motility.The research we did help us and provide a significant foundation and realistic value on explaining the Vibrio cholerae's motility in mucin.
