| Seabuckthorn(Hippophae rhamnoides L.)is a deciduous shrub,which is one of the representative economic crops in Western china.Seabuckthorn is a medicinal and edible plant.Its biological resources are abundant and diverse.Seabuckthorn is a kind of edible and medicinal plant,its stems,leaves,roots and fruits,especially seabuckthorn fruit are rich in bioactive substances and nutrients.As a kind of excellent natural plant protein,seabuckthorn seed protein is rich in content.The protein extracted from seabuckthorn seeds as a kind of excellent natural vegetable protein,rich in amino acids,containing the human body needs all the essential amino acids,can be used as a good plant protein resources.In this paper,Hippophae rhamnoides seed protein was prepared by acid precipitation and alkali extraction.The experimental model of type 2 diabetes mellitus was induced by high fat diet combined with streptozotocin(STZ).For lavage treatment in mice using seabuckthorn protein 4 weeks,probe into its in mice body weight(BW),fasting blood glucose(FBG),blood lipid levels and the influence of liver injury,used to study the seabuckthorn proteins fall hematic fat action on diabetes mice;Testing various periods of mice feces pH value and the number of changes in the intestinal flora to explore the effects of seabuckthorn protein on intestinal microflora and intestinal environment of diabetic mice;At the same time,semi quantitative PCR(RT-PCR)was used to study the effect of mRNA on the expression of fatty acid metabolism,preliminary exploring seabuckthorn seed protein improve diabetes lipid transport mechanism.16 S rDNA sequencing technology was used to separate and classify the whole genome of mice feces,and to investigate the dynamic changes of intestinal microflora in the diabetic mice.Experimental results show that,using the alkali acid sinking process preparation of seabuckthorn seed protein,its content is 78.6%,the total extraction yield of 37.4%.Animal experiment results show that: seabuckthorn protein treatment groups compared with model group mice body weight,its fasting blood glucose,total triglyceride(TG),total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C)were significantly lower than that of diabetic group(p< 0.05).In addition,the results of RT-PCR showed seabuckthorn seed protein make mice liver tissue gene en-hancerbinding protein alpha(CEBP α),Fatty acid synthase(FAS),Acetyl CoA carboxylase(ACC1),Carnitine palmitoyltransferase(CPT1)expression increased,reducing insulin resistance,improve the symptoms of diabetic mice.The cultivation of microorganisms,seabuckthorn protein treatment groups of Bifidobacterium,Lactobacillus and Bacteroides than in the diabetes group were significantly increased,Clostridium coccoides decreased;The results of cluster analysis also showed that the the intestinal flora’s similarity was higher between seabuckthorn protein treatment groups and the normal group,which was about 77%.The results indicated that the seabuckthorn seed protein could improve hyperglycemia,hyperlipidemia and regulate the intestinal microflora of type 2diabetes mellitus.The results showed that seabuckthorn seed protein could improve the metabolism of fatty acids.The results of PCR-DGGE analysis showed that seabuckthorn seed protein could significantly improve the intestinal microflora environment,regulate the dynamic balance and improve the symptoms of type 2diabetes mellitus. |
