| Liver is the main organs of lipid metabolism and important places,fatty liver is one of the common liver disease,the main reason is the accumulation of excessive accumulation of liver cells,cell fattening serious,leading to the formation of fat metabolism disorders.It has been detected that Isoflavones of soybean have a significant therapeutic effect on experimental fatty liver in rats.Isoflavones of soybean are a natural secondary metabolites derived from the roots of legumes,which have significant anti-cancer,antibacterial,antioxidant effects.Genistein(Genistein,Gen.)plays an important role in the activity of soybean isoflavones.It is found that genistein is mainly regulated by oxidase activator receptor(PPARs)and metabolism in lipid metabolism Enzyme expression and regulation of activity play an important physiological function.Objective: To investigate the mechanism of triglyceride regulation of lipid metabolism and To study the regulation of expression of Gen in PPARs,LXR,Adipose Triglyceride lipase(ATGL)and hormone-sensitive triglyceride lipase(HSL).To compare the effects of genistein and PPAR agonists on cell viability and to explore the regulation of human tumor necrosis factor TNF-α and Leptin in adipogenic cells,and to investigate the effects of genistein on the health care,clinical pharmacology,lipid-regulating liver and other aspects to provide theoretical basis and experimental basis.Methods: HepG2,Hep3 b and 3T3-L1 cells were cultured in vitro.The cells were divided into blank group,model group,Gen group and the PPAR group.MTT was used to determine the cell viability at different concentrations of oleic acid(OA).Cell morphology was observed.The optimal concentration of OA was determined and the cell fatification model was established.The cells were stained with Annexin V / PI double staining method.The apoptosis of cells was detected by flow cytometry.On the basis of MTT test,the effects of Gen,PPAR agonists and inhibitors on the regulation of lipid metabolism were studied.Four kinds of lipidized cells were treated with Gen,PPAR activator and inhibitor.The expression of mRNA in ATGL,HSL,PPARα,PPARγ and LXR were detaermined by real-time fluorescence quantitative PCR..The expression of mRNA was detected by real-time quantitative PCR.The levels of TNF-α and Leptin in four kinds of liposomal cell cultures were detected by enzyme-linked immunosorbent assay(ELISA).Results:(1)Cell viability test result of MTT showed that when the concentration ofoleic acid was higher than 1mM,the cell viability decreased rapidly and some cells were floating and necrotic.When the concentration was lower than 1mM,the cell viability was similar and the cell morphology changed obviously.(2)Test results of Elicited cytology show that genistein has a protective effect on cells,it can improve cell activity.(3)Gen can up-regulate the mRNA expression of PPARs and LXR,and the regulation of PPARs was significantly higher than that of LXR,and the expression of HSL and ATGL mRNA was also increased.(4)Gen can regulate the concentration of TNF-α and Leptin in a dose-dependent manner.Conclusion: Genes and PPAR agonists can regulate the cell activity and improve the TG metabolism of cells by using 1mM concentration of OA to induce the cell fatification model.Conclusion:1mM OA is suitable for the establishment of cell fatification induction model,Gen and PPAR agonists can regulate the lipid metabolism of cells,regulate cell TG content.Gen can regulate the expression of ATGL and HSL by metabolic nuclear receptor pathway.TNF-α and Leptin can significantly reduce the expression of ATGL mRNA.Gen is to inhibit the secretion of TNF-α and Leptin by regulating PPARs pathway and regulate the production of lipid metabolism lipid-lowering effect,has important physiological significance. |
