Lung cancer is one of the most common causes of death in the world.The reason is that the lung cancer is a very complex tumor,and the malignant sign of lung cancer is distant metastasis.Therefore,the metastasis of lung cancer is still a major problem.With the awareness of lung cancer metastasis,Traditional Chinese Medicine plays an increasingly important role in the treatment of lung cancer metastasis.Paris polyphylla saponin as the main component of Rhizoma Paridis,has favorableanticancer effect,found in previous experiments,it also has good anti lung cancer metastasis effect.This paper successfully established C57BL/6 mice Lewis pulmonaryadenocarcinoma model.Weight,diet and water index is adopted to assess the quality of life of Lewismice.RPS inhibited lung cancer metastasis effectby anatomical observation,assessment of tumor weight and pathological examination.PCR analyzedlung cancer metastasis process aboutangiogenesis,lymphangiogenesis,adhesion,invasion and nuclear factor related gene.Cytokine antibody microarray estimated lung cancer metastasis related inflammatory cytokines and chemokines.Antioxidant enzyme activities were detected.Meanwhile,cytokines and oxidative stress stimulatedlung cancer metastasis downstream signal pathways ofproteins,the proteins were detected by Western blot.Lewis pulmonary adenoma mice were successfully established to analyze the pathways involved in RPS intervening tumor formation and progression.As a result,RPSinhibited levels of cytokines or receptors such as VEGFD,VEGFR3,RAGE,IL6R,IL17BR and CXCL16,also raised the content of SOD and CAT enzymes,cytokines andoxidative stress were regardedas the initiators induced tumor cell proliferation,adhesion,angiogenesis and invasion.RPS Moreover,RPS inhibited the aberrantly active NF-?B,andphosphorylation of PI3K/Akt and MAPK(including p38,Erk1/2 and JNK)signalingpathways.Soon after,RPS changed mRNA expression of nuclear factors containing NF-?B,HIF-1 A,STAT3 and Jun,and consequentiallysuppressedthe expression of angiogenesis,lymphangiogenesis,adhesion,inflammation and invasion enzymes.The against lung cancer metastasis study found single index(invasion of enzyme and invasion of enzyme inhibitor),it can't fully reflect the traditional Chinese medicine effectiveness,by comprehensive consideration of the quality of life of patients with cancer,angiogenesis,lymphangiogenesis,adhesion,invasion and nuclear factor in lung cancer metastasis process,cytokines and oxidative stress effect induced by lung cancer metastasis downstream signaling pathways,which can fully evaluate the pharmacodynamics of Paris polyphylla saponin.Therefore,this study hopes to prove that RPS is multi targets anti lung cancer metastasis from all aspects,which is beneficial to the application of RPS in the treatment of cancer.
