| Objective : "Cited by the meridians" theory is one of the theoretical system of traditional Chinese medicine prescriptions,some drugs have a special affinity on body certain organs and meridians,when used in combination with other drugs,it has mediated the distribution of other drugs to its attribution of organs the meridians.It can be inferred that guiding medicals have potential target character.To investigate the effect and mechanism of the anti-renal fibrosis property’ radix notoginseng and guiding medicals mediated notoginseng on the renal fibrosis in rats with chronic kidney disease.Methods : Based on the theory of cited by the meridians,a total of 120 male SD rats were divided randomly into six groups,which were Nomal control group,Chronic kidney disease group,radix notoginseng group(RN),radix notoginseng plus lung-related meridian drug platycodi group(RNP),radix notoginseng plus renal-related meridian drug cinnamon group(RNC)and radix notoginseng plus guiding medicals down distribution radix cyathula group(RNR).CKD group,RN group,RNP group,RNC group and RNR group were manufactured with Adriamycin plus Adenine.After modeling,NOR group and CKD group were intragastric administrated with normal saline lavage,RN group were intragastric administrated medium dose radix notoginseng,RNP group were intragastric administrated medium dose radix notoginseng plus the same dose platycodi,RNC group were intragastric administrated medium dose radix notoginseng plus the same dose cinnamon and RNR group were intragastric administrated medium dose radix notoginseng plus the same dose radix cyathula.The rats’ 24-hour urinary protein was detected after the second and the fourth week of drug intervention.After the treatment of 4 weeks,rats were sacrificed,the renal function were assessed by the level of the rats’ serum creatinine and blood Urea nitrogen,the histopathologic change of renal tissue were observed by Hematoxylin-eosin staning,the deposition of collagen and the renal interstitial fibrosis were observed by Masson staining,the content of transform growth factor – β in blood and urine were detected by Enzyme-linked immunosorbent assay,the expression of Fibronectin,Laminin in kidney tissue were detected by immunohistochemistry and the expression of TGF – β,ɑ-smooth muscle actin in kidney tissue were detected by Westetn blot analysis.Results: Compared with the NOR group,CKD group’ 24-hour urine protein significantly increased,exhibited renal dysfunction : Serum creatinine(P <0.01)and Blood urea nitrogen(P <0.01)were significantly increased,renal interstitial severe fibrosis manifestation and increased collagen deposition(P<0.05).The content of TGF – β in blood and urine were elevated and the expression of kidney tissues’ ɑ-SMA(P<0.01),TGF –β(P<0.01),Fibronectin,Laminin significantly increased.Compared with the CKD group,administration of RN,RNP,RNC,RNR significantly decreased Chronic kidney disease rat model’ 24-hour urine protein,alleviated the renal impairment : Serum creatinine(P <0.01)and Blood urea nitrogen(P <0.01)significantly decreased,the deposition of collagen decreased(P<0.05),renal interstitial fibrosis was reduced to varying degrees.The content of TGF – β in blood and urine reduced and the expression of kidney tissues’ ɑ-SMA(P<0.05),TGF-β(P<0.05),Fibronectin,Laminin was downregulated.Moreover,more effective role in antifibrotic was observed in RNC group and RNR group when compared with that of RN group and RNP group.Conclusion : In summary,guiding medicals enhanced radix notoginseng antifibrotic effect in various degrees,retard the progression of fibrotic renal diseases induced by Adriamycin plus Adenine,the mechanism of which maybe related with the inhibition of TGF –β signal transduction pathway.It was revealed that guiding medicals to renal-related Meridian and down distribution have potential renal tissue specific selectivity and targeting,which could be a potential target for ameliorating chronic kidney disease. |
