Objective: To observe the effect of Shuyu Wan on the expression of tumor growth curve,spleen index,IL-2,IL-10,TNF-α and IFN-γ in post-chemotherapy breast cancer mice,and to discuss its immunoregulation function and possible action mechanisms Method: A total of 32 healthy female 6-week BALB/c mice weighting 20±2g were included in this study.Then 24 mice were randomly selected to give left abdominal subcutaneous injection of 0.1ml 4T1 breast cancer cell suspension.After the successful model establishment,mice were divided into model group,chemotherapy group,chemotherapy + TCM group and control group.One intraperitoneal injection of docetaxel(6.28 mg/kg)was provided for chemotherapy group and chemotherapy + TCM group,and then 0.3ml of Shuyu Wan l gavage therapy was given to chemotherapy + TCM group from the morning of the very second day for 21 days in a row in order to observe the behavioral change among groups.The mice were sacrificed after treatment,and the serum levels of IL-2,IL-10,TNF-α and IFN-γ among groups were measured via ELISA.The spleen index and tumor growth curve in mice groups were calculated and drawn.Result: After treatment,Comparing to control group,levels of IL-2,IFN-γ and TNF-α in model group were significantly lowered and IL-10 level was significantly elevated in model group.The difference was statistically significant(P<0.001 or P<O.O5);comparing to model group,IL-2 and IFN-γ in chemotherapy group were significantly reduced,TNF-α and IL-10 were significantly elevated,and the difference was statistically significant(P<0.001 or P<0.01);comparing to CHE group,IL-2 and IFN-γ in chemotherapy + TCM group were significantly elevated,IL-10 was significantly reduced,and the difference was statistically significant(P<0.05),while TNF-α was lowered with no statistical significance(p>0.05).Comparing to model group,the tumor growth was slower and the size was smaller in chemotherapy group and chemotherapy + TCM group with statistical significance(P<0.05);comparing to chemotherapy group,the tumor growth was slower in chemotherapy + TCM group with no statistical significance(P>0.05).;comparing to control group,spleen index in model group was significantly reduced with statistical significance(P≤0.001);comparing to model group,spleen index in chemotherapy group and chemotherapy + TCM group was significantly reduced with statistical significance(P<0.001);comparing to chemotherapy + TCM group,spleen index in chemotherapy group was significantly lowered with statistical significance(P<0.001).Conclusion: The immunity in breast cancer mice was reduced than normal mice,which was aggravated by chemotherapy.Shuyuwan improved the immunity in post-chemotherapy breast cancer mice,and it might be related the the elevation of serum IL-2 and IFN-γ and reduction of IL-10.
