Objective To explore the effect of Resveratrol(Resv)on the proliferation and angiogenesis of human umbilical vein endothelial cells(HUVEC)with the possible mechanismsMethods The HUVEC was treated with LPS and Resv,the proliferation of HUVEC was tested by MTT.The effect of Resv on the ability of angiogenesis of HUVEC was observed by in vitro angiogenesis assay.The ability of migration was tested by Wound healing assay.Nitric oxide(NO)level was measured by nitrate reduction method.The levels of CXCR4?VEGF?MMP-2?COX-2 and iNOS mRNA were measured by Reverse Transcription Polymerase Chain Reaction(RT-PCR).The phosphorylation level of ERK and JAK2 protein were measured by Western Blot.Results LPS promoted the proliferation of HUVEC at a concentration of 10 ng/ml.LPS could significantly promote the ablity of angiogenesis,migration and release NO of HUVEC.LPS also up regulated the protein of p-ERK,p-JAK2 and the mRNA of CXCR4,VEGF,MMP-2,COX-2,iNOS.Conclusion Resveratrol could inhibit the proliferation,migration,invasion of gastric cancer cell.The mechanism may be related with decreasing the expression of p-ERK,p-JAK2,CXCR4,VEGF,MMP-2,COX-2 and iNOS.
