Objective: To evaluate the association of TRAF6 gene polymorphisms and the expression level of TRAF6 mRNA,inflammatory cytokines,and magnetic resonance imaging(MRI)signs of ischemic stroke(IS).Methods: A case-control design was used in this study and a total of 94 controls and 65 IS cases,which consisted of 35 Fengtanyuzu syndrome cases and 30 Qixuxueyu syndrome cases were included.The two single nucleotide polymorphisms(SNPs)of the TRAF6 gene,rs5030411 and rs5030416,were genotyped with the Sequenom MassARRAY iPLEX platform.Real-time PCR(RT-PCR)method was used to determine the TRAF6 mRNA expression and enzyme-linked immuno sorbent assay(ELISA)method was used to detect the inflammatory cytokines levels in serum.The MRI signs of all IS cases were observed.The PLINK software and the SPSS version 16.0.software were used for statistical analysis.Results:1.Association between the expression level of TRAF6 gene and syndrome of traditional Chinese medicine : Compared with control group,the TRAF6 mRNA expression level of IS patients with Fengtanyuzu syndrome and Qixuxueyu syndrome rise significantly(P<0.050).2.The correlation analysis of TRAF6 gene polymorphism and TRAF6 mRNA expression levels: No significant difference was observed in the expression of TRAF6 mRNA among different rs5030411 and rs5030416 genotypes carriers in both IS patients with Fengtanyuzu syndrome and Qixuxueyu syndrome(all P>0.050).3.The association analysis of TRAF6 gene polymorphisms and inflammatory cytokines: In IS patients with Fengtanyuzu syndrome,the CT genotype carriers have a higher level of IL-1??IL-6 than non-CT genotype carriers.4.The association analysis of TRAF6 gene polymorphisms and MRI signs: In both Fengtanyuzu syndrome and Qixuxueyu syndrome IS patients,no statistically significant association was observed between rs5030411 and rs5030416 genotypes carriers and MRI signs,including number of infarction lesions,infarction size and site of apoplexy(all P>0.050).Conclusion: TRAF6 gene might be associated with the occurrence of IS,and rs5030411 polymorphism might be involved in the inflammatory reaction of IS cases with Fengtanyuzu syndrome.Rs5030411 polymorphism was not associatied with the TRAF6 mRNA expression and MRI signs of both Fengtanyuzu syndrome and Qixuxueyu syndrome IS cases.Besides,rs5030416 polymorphism was not associatied with the expression of TRAF6 mRNA,inflammatory cytokines,and MRI signs of IS cases with Fengtanyuzu syndrome and Qixuxueyu syndrome.
