| Objective: Using the technology of 1HNMR,the small molecule metabolites in rabbit knee osteoarthritis model were detected.By making a comparison of multiple groups of knee osteoarthritis in rabbit serum metabonomics data,this experiment explored the serum metabolome of knee osteoarthritis characteristics and metabolic markers,probed the mechanism of serum metabolic characteristics and roubin manipulation in the treatment of knee osteoarthritis,provided theoretical support for the improvement of knee osteoarthritis clinical diagnosis scholarship.Methods: 52 6-month-year old rabbits were chosen in this experiment,each rabbit weight about 2.5-3kg(male or female).Using random number table method,the experiment divided the rabbits into the manipulation group,the control group,the sham group and the normal group,13 in each groupthe.All the rabbits were breed apart in the same conditions.Both the manipulation group and control group were given ligation and cut off the right lower limb inferior gluteal vein,femoral vein and great saphenous vein,causing the right lower limb intraosseous hypertension model,sham group exposure related vein without ligation and cut off.The manipulation group for roubin manipulation were given treatment for 5 weeks,a total of 17 times.The control group,sham group and normal group were not treated.Blood samples were collected from the rabbit ear vein at the end of the 1 week and the 12 week.Through the 1HNMR technology,rabbit serum metabolites were analyzed.Using PLS-DA and PCA methods,this paper probed these problems,and through the VIP map,found the potential biomarkers in the serum.After twelfth weeks,the cartilage of the medial tibial plateau of the right lower limb was examined and scored.Results: 1.The Mankin score of cartilage tissue of animal model: the control group had moderate degeneration of articular cartilage,P<0.05.The articular cartilage of the manipulative group and the sham model group were slightly degenerated,which was between the normal group and the control group,P<0.05.There was no significant difference between the manipulation group and the sham group.2.At the first weekend,manipulation group,model group,sham model group Serum specimen of glucose,lactate,pyruvate content was significantly lower than that in normal group,P<0.05.The reason was the stable model of postoperative wound recovery,postoperative recovery of energy consumption increased.3.After twelfth weeks,the serum glucose level in the manipulation group was significantly lower than that in the other three groups,P<0.05.The serum glucose level in the model group was significantly lower than that in the normal group,P<0.05.At the end of the twelfth manipulation group,model group,serum lactate pyruvate was significantly higher than normal group,P<0.05.Because the manipulation group,model group limb hypoxia,glucose glycolysis increased;roubin manipulation was improved to some extent manipulation group rabbit three carboxylic acid cycle.The concentration of glycine in the manipulation group was significantly lower than that in the sham group,P<0.05.Because roubin manipulation increased to a certain extent,the glycine consumption and chondrocyte metabolism active synthesis of cartilage matrix increased.The consumption of glycine may also be related to its anti-inflammatory and protective effects on energy depleted cells.4.In the first week,the content of serum glucose was higher than that of the control group,the model group and the sham group,P<0.05.In the first week,the manipulation group,model group,sham group,serum lactate pyruvate and urea were less than twelfth weeks,P<0.05.After the model operation,the three groups of serum glucose anaerobic glycolysis and amino acid metabolism increased.Conclusion: In this study,the use of surgical ligation of animal related venous modeling was successful.The formation of animal KOA may be associated with insufficient oxygen supply to the knee joint,increased glucose anaerobic glycolysis,and abnormal metabolism of three carboxylic acids.By improving the body three carboxylic acid cycle,enhancing chondrocyte metabolic activity,promoting chondrocyte proliferation and progressing KOA,Roubin manipulation achieved the purpose of treatment.The increased consumption of Rabbit serum glycine after treatment may related to the enhanced chondrocyte metabolism and the activity involved in the synthesis of cartilage matrix.Roubin manipulation may promote the anti-inflammatory effect of glycine,glycine to protect the chondrocyte energy metabolism disorder in joint,and finally increase the survival ability of articular cartilage cells. |
