Objective Proteomics are applied to screen out specific biomarkers in the salivary of colorectal cancer(CRC)and different syndromes of CRC like spleen deficiency syndrome and damp heat syndrome,in order to obtain a simple,high sensitivity and specificity of diagnostic method for colorectal cancer and its syndromes.Method Using isobaric tags for relative and absolute quantification(iTRAQ)quantitative proteomic technology,screens out different saliva proteins among normal people(45 cases),colorectal cancer with spleen deficiency syndrome(9 cases)and colorectal cancer with damp heat syndrome(18 cases).Then find out the different saliva proteins by using Excel software,the ratio of higher protein is greater than or equal to 1.5,and the ratio of lower protein is less than or equal to 0.667.And applying the Gene Ontology(GO)database to find out the Biological Process(BP),Moleculer Function(MF),Cellular Component(CC)and path analysis of the different proteins.Results 1.In the comparison beteen colorectal cancer with spleen deficiency syndrome and normal people,it screened out 202 proteins which were greater than or equal to 1.5 and 176 proteins which were less than or equal to 0.667.2.In the comparison beteen colorectal cancer with damp heat syndrome and normal people,it screened out 270 proteins which were greater than or equal to 1.5 and 223 proteins which were less than or equal to 0.667.3.In the comparison beteen colorectal cancer with spleen deficiency syndrome and damp heat syndrome,there are the same 45 proteins and 157 special proteins of damp heat syndrome and 225 special proteins of spleen deficiency syndrome,which all the proteins were greater than or equal to 1.5.And there are the same 34 proteins and 142 special proteins of damp heat syndrome and 189 special proteins of spleen deficiency syndrome,which all the proteins are less than or equal to 0.667.Conclusion 1.It screened out some specific molecular markers for differential diagnosis of colorectal cancer with damp heat syndrome based on iTRAQ,a method of quantitative proteomic technology.2.It screened out some specific molecular markers for differential diagnosis of colorectal cancer with spleen deficiency syndrome based on iTRAQ,a method of quantitative proteomic technology.3.It is preliminarily obtained that colorectal cancer with different Traditional Chinese Medicine(TCM)syndrome types (such as spleen deficiency syndrome and damp heat syndrome)had different expression proteins in the biological process,moleculer function,and cellular component based on the iTRAQ technology.
