| ObjectiveTumor microenvironment is one of the most important factors in the development of tumor.Tumor associated fibroblasts,as the most important component of tumor microenvironment,participate in the development,invasion and metastasis of tumor.Through the study of colorectal cancer in rats compared with cold or heat syndrome type two startup tumor associated differences in fiber cells,observe the colorectal tumor tissues of rats related to expression of fibroblasts of three representative markers FAP,fsp-1 and PDGFR,to find the difference mechanism of different syndromes in rats the tumor may exist.MethodsThe rats were randomly divided into model group,cold syndrome and heat syndrome model group and blank group(male and female),first through 5 weeks,to establish the control model and blank with cold or heat syndrome;then making model of colorectal cancer in 12 weeks,and from the cold syndrome group and heat syndrome model group draw half the rats were used as cold syndrome and heat syndrome of traditional Chinese medicine group,Chinese medicine group,giving extra anti Zuojinwan and Zuojinwan Chinese medicine disproof.From eighteenth weeks,the specimens were collected every three weeks to observe the incidence of colorectal cancer and the expression of FAP,fsp-1 and PDGFR in each group.ResultCold and heat syndrome model rats appear obvious cold or heat syndrome symptoms and signs associated with cold and heat syndrome index ATP and SDH enzyme activity had obvious difference;colorectal cancer model established on this basis,through the pathological detection of large bowel tumor was no significant difference between the model.Group of cold syndrome and heat syndrome,and the traditional Chinese medicine intervention group of the corresponding cancer there is a big difference between the situation with the model group.Finally,immunohistochemistry and PCR results showed that tumor associated fibroblast markers FAP,fsp-1 and PDGFR were significantly different between the model group and the Chinese medicine intervention group,while there was no obvious difference between cold and hot two model group.Conclusion:1.It is feasible to establish the model of colorectal cancer on the basis of cold and heat syndrome.The model reflects the difference between cold and heat,and the induced colorectal cancer model has the characteristics of cold and heat syndrome,which provides an experimental animal model for further study of the mechanism of carcinogenesis.2.Cold syndrome of colorectal cancer with cancer specific progress slow,but also more serious.3.Inhibition of Zuojinwan and Fanzuojin Wan on colorectal cancer is mainly reflected in the early stage of tumor,and the inhibitory effect is closely related to its proportion. |
