| Objective: The two subunits Ebi3 and IL-12p35 for the detection of IL-35 expression of in oral leukoplakia and oral squamous cell carcinoma,to explore the role of IL-35 in the process of oral leukoplakia canceration,providing new reference indicators for the early diagnosis and prognosis of oral squamous cell carcinoma.Methods: 1: 32 cases of oral leukoplakia,34 cases of oral squamous cell carcinoma and 15 cases of normal oral mucosa,which all tissues been embedded by paraffin.The two subunits expression of Ebi3 and IL-12p35 of IL-35 in protein level was detected by immunohistochemical technique,and analyzing these changes and significance in the process of carcinogenesis in oral mucosa.2.31 cases of oral squamous cell carcinoma and 20 cases of normal oral mucosa.Q-PCR was used to detect the gene expression of p35 and Ebi3 in OSCC lesions,analyzing the relationship between these factors and the clinical pathological features of OSCC.Results: 1.There was no expression of Ebi3 in normal oral mucosa,but Ebi3 protein began to appear positive expression of simple hyperplasia tissues in oral leukoplakia;the positive expression of Ebi3 in epithelial hyperplasia tissues was 31.25%(10/32),and in OSCC was 58.82%(20/34),there were statistically significant differences between the two groups(χ2 =5.055,P= 0.025).The Ebi3 positive expression rate and expression intensity increased with the increase of the degree of epithelial dysplasia,there was a positive correlation between the two groups(r =0.409,P﹤0.05).There was a relationship between the expression of Ebi3 and OSCC lymph node metastasis(P < 0.05),but not with gender,age,the degree of differentiation and clinical stage of tumor(P >0.05).2.There was no expression of IL-12p35 in normal oral mucosa,but IL-12p35 protein began to appear positive expression of simple hyperplasia tissues in oral leukoplakia;the positive expression of IL-12p35 in epithelial hyperplasia tissueswas 53.12%(17/32),and in OSCC was 67.65%(23/34),which were higher than the control groups,the difference was statistically significant(P < 0.05);but there was no significant differences between the two groups(P>0.05).The IL-12p35 positive expression rate and expression intensity increased with the increase of the degree of epithelial dysplasia,there was a positive correlation between the two groups(r=0.354,P﹤0.05).There was a relationship between the expression of IL-12p35 and OSCC lymph node metastasis(P < 0.05),but not with gender,age,the degree of differentiation and clinical stage of tumor(P>0.05).3.The expression of Ebi3 and p35 mRNA in patients with oral squamous cell carcinoma was significantly higher than that in control groups(Z1=-3.119,P1=0.002;Z2=-2.116,P2=0.034),there was a statistically significant difference and a relationship with the lymph node metastasis in patients with carcinoma(P <0.05).Conclusions: 1.Ebi3,IL-12p35 may be an early event in carcinogenesis of oral mucosa 2.The expression of IL-12p35,Ebi3 protein increased with the increase of the degree of malignant tissue,suggesting that the process of oral squamous cell carcinoma in the development process of IL-35 immunosuppression 3.IL-35 may be used as a new indicator for the prognosis of oral squamous cell carcinoma. |
