In this study,we concentrated and extracted triterpene acids from Seabuckthorn leaves,to explore the crude extracts of seabuckthorn leaves has the function of anti-diabetics.Establish rat model of type 2 diabetes mellitus by feeding high-fat diets in conjunetion with the injection of small-volume of alloxan.Intervene with preventive administration of crude extracts of seabuckthorn leaves,observe the actions of seabuckthorn leaves crude extracts against rat model of type 2 diabetes mellitus.To provide reference for the treatment of diabetics and lay a foundation for the development and industrial production of glucose-lowering health care products of hippophae rhamnoides.During the experiment,by using boiled-impurity of low alcohol-ethanol extracts a series of steps to get the triterpene acids extracts of seabuckthorn leaves,Corosolic acid content is detected as the index and establishing HPLC method to determine corosolic acid content.All groups,except control group,had been modeled by injecting alloxan after having been fed with high fat diet for continuous 33 days,while giving the low,medium and high three dose groups sea buckthorn leaves crude extracts.After the test,to observe the changes of rats in body weight,fasting glucose and glucose tolerance,blood lipids and changes of serum insulin and c-peptide.The experiment on the seventh,fourteenth,twenty-one and twenty-eight day,drew blood by pricking skin of tail then processed blood smear.The microslides were satained by Wright-Giemsa then photographed,each kind of cells was observed identified and counted.The results show that the content of corosolic acid in sea buckthorn leaves was respectively 2.14%.The high dose could alleviate weight loss.The fasting blood glucose was declined by medium and high doses,they could impaired glucose tolerance and Insulin resistance.In aspect of blood-lipid regulation,medium and high dose could improve triglyceride content and index of high-density lipoprotein of SD rats.The results had statistical significance.Medium dose declind cholesterol content of SD rats.Medium and high dose groups improved low density lipoprotein(LDL-C)content,but there was no significant.From observation of blood cells,it indicated that medium and high doses improved the percentage of neutrophile granulocyte and lymphocyte.medium and high doses reduced the ratio of neutrophils and lymphocytes.medium and high dose groups improved the neutrophil hypersegmentation and lymphocyte chromatin condensation.In model experiment,it suggested that the triterpene acids crude extracts of seabuckthorn leaves regulated blood glucose and blood lipid effectively,it could improve insulin resistance and inflammatory state.It speculated that the triterpene acids crude extracts of seabuckthorn leaves to improve type 2 diabetes may have a certain relationship with the increase islet cell protection and immunity.In addition,by HPLC determining corosolic acid content were accurate,reproducible and can be used for the determination and quality control of sea buckthorn leaf crude extract.
